Primula veris / Echte sleutelbloem

De Sleutelbloem, Primula officinalis of Primula veris, komt in ons land in vochtige weilanden en in bossen nog wel voor. Het is vooral in de Ardennen dat we de gele bloemen volop zien bloeien. De Sleutelbloem, vroeger ook wel hemelsleutel genoemd, is een der eerste voorjaarsbloemen, vandaar ook de geslachtsnaam Primula, van het Latijnse primus, de eerste.

In de antieke geschriften is de gewone sleutelbloem blijkbaar niet terug te vinden en waarschijnlijk was zij in de oudheid niet bekend, mogelijk omdat zij in Griekenland en in Italië weinig voorkomt. Pas bij Hildegard von Bingen, abdis van het klooster op de St. Ruprechtsberg bij Bingen (1098—1179) vinden wij in haar werk „Physica" de naam „Hymelslozel" vermeld. Zij gebruikte deze plant tegen melancholie, hoofdpijn en verlammingen, In de kruidenboeken van de 16e eeuw verschijnt de Primula officinalis onder verschil­lende namen, zooals Herba Paralysis (bij Otto Brunfels), Verbasculum odoratum (Fuchs), Arthritica (Gesner) enz. Laatstgenoemde benaming wijst op het gebruik van de plant tegen arthritis en jicht. Vooral in de wijn getrokken Sleutelbloemen ston­den in die tijd in hoog aanzien.

Sleutelbloem bij Bock en van Beverwijck

De kruidkundige Hieronymus Bock bericht in zijn kruidenboek (1731) over de Sleutelbloem, die bij hem Betonica alba, Witte Betonie noemt, dat de in wijn ge­trokken bloemen als schoonheidsmiddel dienen om vlekken en puistjes uit het gelaat te verwijderen. De Dordtse geneesheer Johan van Beverwyck schrijft in zijn „Schat der Ongesontheydt ofte Geneeskonste van de Sieckten" (1656) : „Sleutelbloemen, die oock om de gehelyckenisse van haer bla­deren Witte Betonye ghenoemt werden, werden vordelyck ghebruyckt in alle koude ghebreken der Herssenen en Zenuwen, insonderheyt met Salye (Salie) en Maryoleyne (Marjolein) ge­kookt. Welcken drank oock bequaem is voor de gene, die bevende Leden hebben, of geraeckt en beroert zijn, 't welck de oorsaeck is, dat de Sleutelbloemen in 't Latijn oock Herba Paralysis ghenoemt zijn".

Volkse verhalen over de sleutelbloem

Als eerste lentebode moet, volgens het volksgeloof, de Sleutelbloem bijzondere geneeskracht bezit­ten en kon zij zelfs voorbehoedend werken; zo worden in het vroegere Pruisen drie Sleutelbloemen ingeslikt als een voorbe­hoedmiddel tegen de koorts en de Roethenen in Boekowina deden dat ook om zich tegen klierziekte te beschermen. Onder bepaalde omstandigheden konden Sleutelbloemen echter ook zeer schadelijk werken en in vele streken meende men, dat het in huis halen van deze voorjaarsbloemen ongeluk ­bracht. In Engeland en Frankrijk werd verteld, dat de kuikens niet uit de eieren kunnen kruipen, als men Sleutel­bloemen in huis heeft gebracht. In Beieren heerste de volksmening, dat men uit de lengte van de Sleutelbloemen de hoedanigheid van de graanoogst kon aflezen. Hadden de Sleutelbloemen n.l. lange stelen, dan zou de gerst dat jaar hoog worden, bleven ze kort, dan zou ook de gerst kort zijn.

De Sleutelbloem geldt verder, vooral in Frankrijk, als een orakel voor trouwlustige meisjes. Meerdere bloe­men in een glas water kregen de naam van een aanwezig meisje. De bloemen, die dan rechtop in het water bleven staan, brachten het betreffende meisje geluk, de bloemen die snel verwelkten betekende ongeluk. In Bretagne zou het meisje, dat een Sleutelbloem met zeven (in plaats van vijf) kroonbladen vindt, nog in het zelfde jaar een man krijgen. (H. Marzell).

In een volkssage werd de Sleutelbloem gebruikt om in toverbergen de schatten, die er verborgen waren aan te wijzen. Dit steunt op de volksetymo­logie dat sleutelvormige bloemen de deuren van onderaardse schatplaatsen kunnen openen.

Sleutelbloem in apotekersboeken

In de apotekersboeken van 19de en zelfs 20ste eeuw werden vooral de sleutelbloemwortels als geneeskrachtig beschreven. De 'radix primulae' werden voorgeschreven als expectorans, dus vooral gebruikt om taai slijm uit de luchtwegen te verwijderen. Maar ook als pijnstillend en ontstekingswerend middel waren ze bekend. De wortels bevatten aspirine-achtige stoffen, die je ook kan reuken als je de wortels oogst.

La Primevère commune, Primevère acaule ou Primevère sans tige (Primula vulgaris) est une plante herbacée pérenne de la famille des Primulacées des régions tempérées de l'hémisphère nord.

Elle fleurit au début du printemps (février-avril) sur les talus et dans les prés. Si elle est inscrite sur la liste des espèces végétales protégées en Lorraine, on la rencontre abondamment dans l'Ouest de la France, où elle supplante localement les Primevères officinales (Primula veris). Elle se distingue de cette espèce par des fleurs jaune pâle, à pétales étalés et dont les pédoncules partent tous directement de la base de la plante. Dans les zones où ces deux espèces cohabitent, on peut en rencontrer des hybrides.

C'est d'ailleurs une des parentes de la primevère cultivée avec la primevère élevée (Primula elatior), la primevère officinale (Primula veris) et Primula juliae.

Composition and cardioprotective effects of Primula veris L. solid herbal extract in experimental chronic heart failure.

Background: High interest in chronic heart failure (CHF) is accounted for by its high incidence, poor prognosis, growing number of hospital admissions due to the heart failure relapse, and inadequate treatment. These facts necessitate a search for new pharmacological agents for the CHF correction. Herbal medicinal products appear to be very promising as they have a noticeable therapeutic effect and tend to be more harmless in comparison to the most of synthesized medications.

Purpose: Our aim was to study the composition of the Primula veris L. solid herbal extract (PVSHE) and its effects on the myocardial contractile function in animals with experimental CHF.

Study design: The study design involved the identification of the raw material composition of the P. veris L. extract. For the experimental part of our research, we used the model of CHF to elucidate the cardioprotective properties of PVSHE.

Methods: The active extract constituents were isolated by thin-layer chromatography and column chromatography; the extract components were identified by high-performance liquid chromatography, ultraviolet spectroscopy (UVS), and nuclear magnetic resonance spectroscopy (NMRS). To model CHF, L-isoproterenol at a dose of 2.5 mg/kg was intraperitoneally injected to the experimental rats twice a day for 21 days. Cardiac output was assessed with the loading test, adrenoreactivity test, and maximum isometric loading test; CHF markers adrenomedullin and copeptin were detected in blood plasma with ELISA kit for adrenomedullin and copeptin (Coud-Clone Corp., USA).

Results: P. veris L. solid herbal extract contains flavonoid aglycons (apigenin, quercetine, kaemferol), flavonoid glycosides (cinarozid, rutin, hyperozid), as well as polymethoxylated flavonoids acting as chemotaxonomic markers for the genus Primula (8-methoxy-flavone; 3',4'methylenedioxy-5'-methoxyflavone). The substance 3',4'methylenedioxy-5'-methoxyflavone has been isolated from the primrose herb for the first time. We showed that the PVSHE has a cardioprotective effect when it was administered at a dose of 30 mg/kg in the experimental CHF, as evidenced by a lower number of animal death, lower level of CHF markers in the blood plasma of the experimental animals, the higher increase in rate of myocardial contraction and relaxation, the higher level of left ventricular pressure (LVP) and of maximum intensity of structural performance (MISP), as compared to the control group.

Conclusion: P. veris L. solid herbal extract contains flavonoid aglycons, flavonoid glycosides, and polymethoxylated flavonoids. The herbal agent increases the myocardial contractility in experimental CHF.

The mode of the expectorant action of Primula saponins is not yet satisfactorily clarified. In literature there is a general agreement that saponins irritate locally the gastric mucosa, which provokes a reflex increase in bronchial secretion, which dilutes the mucus and reduces its viscosity (Hänsel R et al (1994), Boyd EM (1954), Hänsel R et al (2007), ESCOP monographs (2003)). Irritation of mucous membranes in the throat and respiratory tract by saponins may also cause an increase in bronchial secretion, and the surface-tension lowering action of saponins might help to reduce the viscosity of sputum, making it easier to eject (Hostettman K et al. (1995). Recently a very specific influence on the β2-adrenergic receptors of alveolar cells has been reported for the saponins of Hedera helix, which is used for the same indications like Primula root (Häberlein H et al (2005)). At present it is not known whether these effects are restricted to saponins of Hedera. 

IN VITRO EXPERIMENTS Most of the published in vitro experiments deal with the antiviral, antimycotic and antibacterial activity, which are common properties of saponins independent of their plant source. Wolters B (1966) has compared the antifungal and antibacterial effects of 30 herbal drugs containing saponins. Among these drugs, Primula root extracts belong to the group of extracts with the most pronounced fungistatic or fungicide effects, while the antibacterial effect of Primula root extract is much less. The author regards saponins as possibly important resistance factors of the plants. Tschesche R et al (1965) describe both antifungal and antibacterial (Staphylococcus aureus, Escherichia coli) effects of Primula saponin (Primula elatior). Against various strains of Candida albicans the total saponins isolated from Primula acaulis (= P. vulgaris Huds.) were effective at concentrations of 80 – 97 µg/ml, comparable to nystatin at concentrations of 2-45 µg/ml (Margineanu C et al (1976). The antimycotic effect of these saponins is quantitatively less than that of the typical antimycotics nystatine and stamicine. The aglycones of the saponins of P. vulgaris root are identical with those found in the roots of P. elatior. An unspecified saponin mixture from Primula veris exhibited activity against influenza(A2/Japan 305) virus, producing 89 % inhibition at a concentration of 6,2 µg/ml (Rao GS et al (1974); Büechi S (1996)). Further in vitro experiments: A hexane extract (50 µg/ml) of Primula veris root inhibited COX (cyclooxygenase)-1 and COX-2 by 54 % and 66 % respectively (Lohmann K et al (2000)). Since this finding has only been published as a poster and clinical studies on a possible analgesic effect of Primula root extracts are lacking, this finding is of no clinical relevance with regard to the traditional use of Primula root. Oswiecimska M et al (1975) have described antimitotic activity of saponin fractions and extracts from Primulae radix and other herbal substances by means of the allium test. 5 Not required as per Article 16c(1)(a)(ii) of Directive 2001/83/EC as amended Page 11/13 EMEA 2016 Herre E (1937) has given some data in rats concerning diuretic properties of Primula officinalis (= Primula veris), but there are no more recent investigations which endorse these findings.

IN VIVO EXPERIMENTS Experiments relevant for the proposed indications: In vivo studies (rabbit) on pharmacological / toxicological effects of extracts from Primula flower showed a significant increase in the production of bronchial secretion at the concentrations tested (Chibanzuga G et al. (1984)). The observed effect was in the range of the reference substances bromhexin and acetylcysteine which had been tested simultaneously. Even though these studies had been carried out with extracts from Primula flower, they might be of some interest: The German Commission E has given the same indications to both Primula flowers and Primula root; Primula flowers also contain saponins. An undefined mixture of saponins from Primula root, at a concentration of 1:10,000, increased the ciliary activity of throat epithelium of curarized frog. This effect was assumed to be due to a decrease in surface tension of the mucus. The ciliary activity was less at a concentration of 1:6,000 and ceased at 1:3,000 due to toxic effects (Vogel G (1963)). Further in vivo experiments: An unspecified saponin of Primula root, administered parenterally, inhibited the growth of Walker carcinoma in rats with an ED50 of 40 mg/kg, although this dose was too toxic in relation to the LD50 of 70 mg/kg to be of practical significance (Tschesche R et al (1973)). Sufka KJ et al (2001) tested herbal extracts for their anxiolytic properties in the chick social separation-stress procedure. For Primula veris (plant part not mentioned) no sedative effects were observed, in addition no alteration of stress responses could be detected.

De sleutelbloem (Primula veris).

Zodra de zon in het voorjaar de nog vochtige bodem verwarmt, kan men in het bruine verdorde gras, langs de kant van de weg en aan de zonkant van heggen

en struiken de eerste sleutelbloemen (Primula vut is) ontdekken. Ze hebben een sterkere gele kleur dan de nauw verwante hoge sleutelbloem (Primula eliator). Deze is niet zo van het licht afhanke­lijk; zij komt meer in elzenbosjes en langs beken voor. Vlak tegen de grond gedrukt, ten dele verbor­gen in de reeds in aanleg aanwezige bladrozet, hebben de bloemknoppen de hele winter gerust. Zij ontplooien zich nu heel vlug en, worden daarbij door de snel groeiende steel omhoog gedreven. Als men nauwkeuriger toeziet krijgt men de indruk, dat de bloei bij de Primula op een eigenaardige manier zelfstandig is geworden. Los van de normale plantaardige ontwikkeling – die eerst tot de ontplooïng van de groene plant leidt – snelt hier het bloeiproces zonder relatie met het overige gebeuren vooruit. Reeds op de grond beginnend, door de bladloze steel snel en ver van de nog onontplooide bladeren verwijderd, verloopt het bloeien volgens eigen wetten. Talloze vroeg vlie­gende vliesvleugeligen en vlinders bezoeken de licht geurende klokbloemen, die afzonderlijk min­der opvallend dan bij andere Primulasoorten, hoofdzakelijk door hun aantal opvallen. De bloei is spoedig voorbij. Opvallend is, dat de bladeren en zelfs de vruchtstelen hun groei tot aan de rijping van de nu niet meer hangende, maar rechtopstaande zaaddozen voortzetten en daarbij een veelvoud van hun grootte ten tijde van de bloei bereiken. Na een rustpoos in de zomer ontstaat in de herfst, als de grond weer vochtiger wordt, uit de solide basis van een korte wortelstok en van lange, sterke wortels een nieuwe bladrozet met de aanleg van bloemen voor het komende voorjaar.

De Primula vertoont, zoals alle vertegenwoordigers van deze familie, een nauwe relatie met de vochtige aarde en een sterke behoefte aan licht. Als bijvoorbeeld door bemesting de vegetatie rondom te overvloedig is, zodat de Primula ’s worden overwoekerd, verdwijnen ze na een paar jaren.

Als geneeskrachtige plant is de Primula tegenwoordig zo goed als vergeten. Men gebruikte bloem en wortel vooral bij stoornissen ten gevolge van verkoudheid en slijmvorming in de longen en bronchiën, bij reumatische verschijnselen en stoornissen van de nieren en de blaas. Behalve saponinen en salicylzuurverbindingen zijn door middel van analyse later stoffen in de Primula aangetoond die de reeds lang bekende werkingen ten dele veroorzaken. Het is begrijpelijk dat volgens het beeld dat de plant vertoont, de werking ervan optreedt waar het vloeistofsysteem en de luchtorganisatie in de mens elkaar raken.
