Origanum vulgare / wilde marjolein

Oregano in soorten

Al groeit er in België maar één soort Oregano in de natuur, toch is er veel verwarring over de verschillende variëteiten en vooral over de naam. De Nederlandse botanische naam is Wilde marjolein, de Latijnse Origanum vulgare maar het gedroogde kruid in de keuken wordt dikwijls met Oregano aangeduid, terwijl de soort Origanum majorana ook Marjolein of Majoraan genoemd wordt.

Andere soorten zoals Origanum vulgare ssp. hirtum (Griekse oregano) en Origanum onitis groeien vooral in het Mediterrane gebied en dan vooral in Griekenland en Turkije. Het zijn meestal ook deze sterker geurende soorten die als keukenkruid verkocht worden. Voor de tuin zijn ook de lage Origanum compactum en de geelbladige ‘Aureum’ interessant, deze variëteiten zijn ook in de lage rotstuin te gebruiken.

Een taxonomisch overzicht van het genus Origanum is in 1980 gemaakt door de Nederlandse botanicus Ietswaart, die maar liefst 38 soorten op naam bracht. Daarbij komt nog dat zowel Lippia-, Plectrantus- en Thymussoorten onder de naam Oregano aangeboden worden. Zo is de bekende etherische olie Spaanse oregano plantkundig gezien Coridothymus capitatus. De verwarring ontstaat ook omdat veel lipbloemigen zoals Tijm, Bonekruid en Oregano hun gebruik in de keuken en als medicijn te danken hebben aan dezelfde geurstoffen met een hoog gehalte aan thymol of carvacrol. Ze reuken dus een beetje hetzelfde! De kwaliteit van onze inheemse Wilde marjolein is door zijn mindere geur met minder carvacrol dan ook spijtig genoeg niet zo geschikt als pizzakruid. Beter kun je dan ook de Origanum compactum en vooral de Griekse oregano, Origanum heracleoticum gebruiken, soorten die ook in ons klimaat goed te telen zijn.

Etherische olie van Oregano: thymol en carvacrol

Origanum vulgare ssp. hirtum met veel carvacrol is vooral ontstekingswerend en kan net zoals Tijm bij verkoudheid, hoest en bronchiale aandoeningen gebruikt worden. De zoetgeurende Origanum majorana heeft veel meer een krampwerende en ontspannende werking, het heeft een zogenaamd parasympathisch effect en wordt daardoor toegepast bij vegetatieve spanningen zoals maagkrampen, benauwdheid op de luchtwegen, zenuwachtigheid en zelfs seksuele overspanning.

Etymologie oregano

In de huidige vorm is oregano ‘keukenkruid’ ontleend aan Spaans orégano ‘wilde marjolein’ [14e eeuw; Corominas], uit Latijn orīganum, dat zelf ontleend is aan Grieks orī́ganos, oreíganos, waarvan de verdere herkomst onbekend is; de plant is afkomstig uit Afrika. De oudere vorm origane met als voornaamste betekenis ‘geneeskrachtig kruid’ is wrsch. ontleend via Frans origan ‘id.’ [13e eeuw; TLF]. De oudste vorm orega en de varianten orego, orege ‘artsenijkruid’ moeten ook zijn ontleend via het Iberisch schiereiland, waar varianten bestaan als Catalaans orenga en Gallicisch ourogo. Orego komt als naam van een geneeskrachtig kruid voor tot zeker halverwege de 20e eeuw (WNT).

In de oudheid verklaarde men de Griekse naam pseudo-etymologisch als een samenstelling van óros ‘berg’ en gános ‘glans, schittering, vrolijke aanblik’; de betekenis zou dus ongeveer zijn ‘dat wat schitterend, vrolijk is in de bergen’.

Van de Majoraan werd van oudsher ook een zalf gemaakt met 3 theelepels verpoederde gedroogde Oregano majorana, 1 uur getrokken in 1 eetlepel wijngeest (alcohol 40°), dan vermengen met 2 eetlepels boter, dit alles 5 minuten au bain marie verwarmen, door verbandgaas uitzeven en de zalf is klaar. Dit is als eerste hulpmiddel te gebruiken tegen buikpijn en krampjes bij baby’s, bij verkoudheid om de neus in te wrijven en vooral ook goed om de pijn bij kneuzingen te verzachten.

Misschien zijn juist warmte minnende planten extra geschikt om een ‘warme’ wereld te verwezenlijken. Ik zou zeggen: ‘ Warme schermbloemigen en vurige lipbloemigen verenigt u!’

Ouder onderzoek met etherische olie van oregano

           Dr. Jean Valnet, The practice of Aromatherapy -London Times may 8, 2001

Jean Valnet documenteerde voor het eerst de antiparasitaire eigenschappen van O. vulgare en gebruikte de olie in lage concentraties om rioolwater te desinfecteren. Griekse wetenschappers bevestigde zijn werk in 1995 in een oplossing van o. vulgare van 1:4000 om rioolwater te sterilizeren.

Tests van het U.S. Federal Government gaven aan dat de olie alle 9 bacillen doodden includief Salmonella, Escherichia coli, Listeria, Stafycoccen, Pseudomonas en schimmels. Waarschijnlijk werd deze test uitgevoerd naar aanleiding van claims betreffende de bacillendodende werking van de olie, ter verificatie.

Paul Belaiche testte veertig verschillende e.o.’s om hun effectiviteit tegen een gevarieerd aantal van micro-organismen vast te stellen. Oregano olie bewees hier de meest krachtigste en meest omvattende olie te zijn.

Uit onderzoek van Prof. Harry Preuss M.D. van de olie van Origanum vulgare bleek dat de groei van Stafylococcus bacteriën even effectief werd geremd, als door Streptomycine, Penicilline en Vancomycine. Ook belemmerde de olie de groei van Candida albicans even effectief als Amphotericin en Nystatin.

Onderzoek van Sarer en collegee en gedocumenteerd in Essential oils and Aromatic Plants demonstreerde een opmerkelijk effect van de olie van Origanum vlgare tegen een omvangrijk gebied van microben, waaronder Candida albicans, Staf., Strept., E. coli en Pseudomonas microben.

Onderzoekers van de Universiteit van Tennessee demonstreerden dat de olie van oregano de grootste antibacteriële activiteiten ontplooiden van alle geteste e.o.’s tegen Staf., E. coli en Listeria.

Er is geen natuurlijke of synthetische antibiotica die meer microben dood dan de olie van Origanum vulgare. Er is geen bewijs dat mutante antibiotisch-resistente “super-bugs”ook resistent zijn tegen Origanum vulgare. Er is geen bewijs dat bacteriën kunnen muteren naar een vorm van resistentie tegen Origanum vulgare.

Echter Dost, Oregano, Origanum vulgare

Die ursprünglich im Mittelmeerraum beheimatete Pflanze wird heute weltweit in warmen und gemässigten Breiten angebaut und genutzt. Natürliche Vorkommen sind in fast ganz Europa vorhanden. Oregano bevorzugt warme Standorte auf kalkhaltigem Untergrund. Er besiedelt gerne trockene und lichte Wälder, wie Eichen- und Kiefernwälder oder Schneeheiden-Kiefernwälder. Gebüsche an Weg- und Waldrändern, sonnige Hänge und Hecken zählen ebenso wie Mager- und Trockenrasen zu seinen regelmässigen Wuchsorten.

MERKMALE

Beim Oregano handelt es sich um eine ausdauernde krautige Pflanze, die Wuchshöhen von 20 bis 70 cm erreicht. Charakteristisch ist ihr ausgeprägter herb aromatischer Duft und Geschmack. Aus einem oft etwas holzigen Rhizom (umgangssprachlich „Wurzelstock“) treibt der aufrecht wachsende, vierkantige und von Grund an gabelig verzweigte Stängel. Dieser weist ebenso wie die meist rötlich überlaufenen Zweige eine leichte Behaarung auf.

Die in endständigen oder seitenständigen dicht gedrängten kugeligen Scheinrispen zusammengefassten Teilblütenstände entwickeln zwischen Juli und September zahlreiche Blüten.

CarvacrolOrigani herba - äther. Öl (bis zu 1 %) mit dem Monoterpenphenol Carvacrol als wichtigste Komponente, im weiteren Flavonoide (Naringin, Apigenin und Luteolin), sowie Phenolcarbonsäurederivate. 

Origani aetheroleum - Terpene mit Carvacrol (bis zu 70 %), γ-Terpinen, p-Cymen, α-Pinen, Myrcen, α-Terpinen und Thymol als wichtige Bestandteile. 

ANWENDUNG

Dostenkraut wird bei Erkrankungen und Beschwerden im Bereich der Atemwege, Husten, Bronchialkatarrh, als Expectorans und bei Husten als krampflösendes Mittel, weiterhin bei Erkrankungen und Beschwerden im Bereich des Magen-Darm-Traktes, Blähungen, zur Förderung der Gallenproduktion und der Verdauung sowie als appetitanregendes und krampflösendes Mittel, ferner bei Erkrankungen und Beschwerden in Bereich der Harnwege, Unterleibserkrankungen, schmerzhafter Menstruation, als harntreibendes Mittel sowie bei Rheuma, Skrofulose und als beruhigendes und schweisstreibendes Mittel angewendet. Dostenkraut ist ausserdem in Gurgelwässern und Bädern enthalten. 

Die Wirksamkeit der Droge bei den beanspruchten Anwendungsgebieten ist nicht belegt.

Bewertung der Kommisssion E:

Da die beanspruchten Anwendungsgebiete nicht belegt sind, kann eine therapeutische Anwendung der Droge nicht befürwortet werden.

SONSTIGES

Dost wurde im Mittelalter als wichtige Hexenabwehrpflanze angesehen und sollte vor dem Teufel schützen. Man hielt sie den Hexen unter die Nase, um sie vom Teufel zu lösen. Dost soll das Kraut sein, das Kummer verschwinden lässt, erloschenen Lebensmut wieder aufrichtet und den Menschen fröhlich macht. Aus diesem Grund trägt die Pflanze auch den Namen Wohlgemut. 

Oregano oder Dost wurde als Schutz vor bösen Mächten in den Brautschuh gelegt und in den Brautstrauss eingebunden. In Form von Räucherungen wurde Dost schon in der Antike als dämonenabwehrendes Mittel verwendet. Wenn ein Kind lange nicht zu reden begann, gab man ihm einen Löffel voll Oreganowasser. Gegen Epilepsie liess man den Kranken an mit den Fingern zerdrücktem Dost riechen.

Origanum vulgare / Wild oregano monograph

Wild oregano (Origanum vulgare) belongs to the Lamiaceae family (also known as Labiatae), which also includes rosemary, thyme and mint. Origanum is the classical Greek name derived from the words ‘oros’ (Mountain) and ‘ganos' (Joy), which refer to the plant’s beautiful appearance in the mountains or from ‘origanon’, which means bitter herb. 

Wild oregano (Origanum vulgare), also confusingly named ‘wild marjoram’, is a herb that is rich in the phenol carvacrol (also known as cymophenol), which is characterised by a spicy and bitter taste with a light sensation of heat. This is in contrast to Origanum majorana or sweet marjoram, which is often used in the kitchen but is less rich in therapeutic substances. To distinguish sweet or real marjoram, this article uses the commonly used term ‘wild oregano’ for the bitter tasting (and carvacrol-rich) Origanum vulgare.

The most potent wild oregano varieties grow in mountainous East Mediterranean areas in chalk-rich soils. Carvacrol is the most researched substance and synthetic carvacrol has been used for over fifty years in cleaning agents such as Lysol and Listerine to disinfect work surfaces. 

Modern interest in the original plant substances has been revived again because the resistance of bacteria and fungi against modern remedies such as antibiotics is forcing us to look for new solutions. Many vegetable-based substances turn out to have broad-spectrum therapeutic effects to a lesser or greater extent against bacteria, yeasts, fungi and parasites as well as against viruses. Wild oregano oil seems to be an all-round remedy in this respect. Veterinary research carried out for dairy and poultry farmers points to possibilities for reducing the use of antibiotics by putting wild oregano oil into fodder and forage. The human foods industry is more interested in using wild oregano oil as a natural preservative. Oregano was already used in ancient Egypt to conserve food and to clean wounds. In ancient Greece it was used against headaches, ulcers, lung disease, asthma and spasms. In modern Greece, oregano is used in teas as a folk remedy against colds, sore throats, influenza, stomach ulcers and other stomach and intestinal complaints.

The carvacrol-rich essential oil in wild oregano turns out to have excellent properties as regards versatility and efficiency. The effects depend on the dose; until now every microbe exposed to essential wild oregano oil has been killed or its growth has been slowed.  

Wild oregano oil exhibits a broad anti-microbial effect when used against food pathogens and contains potent anti-oxidants which have been shown during research to have therapeutic value for both the cardio-vascular and nervous systems. Wild oregano acts like a brake on infection and regulates glucose and cholesterol metabolism. In spite of the increasing number of in vivo studies, there has still not been a proper placebo-controlled clinical trial in humans. The rich history and positive anecdotal evidence are also representative for the potential clinical value of wild oregano oil.

Properties:

Constituents

The essential oil in wild oregano varieties principally consists of carvacrol followed by its precursors p-cymene and gamma-terpinene.  These are also precursors for thymol, which is an isomer of carvacrol, but which represents only a small percentage of the wild oregano oil that originates in the East Mediterranean mountains in comparison with carvacrol. Variations in concentration occur naturally. Semi-finished products made from the essential oil can be standardised for further processing.

Active effects:

Reduction of stress in cells

In vitro research shows that, up to now, carvacrol is the most potent modulator of TRP channels in mammal cells. Other substances such as cinnamaldehyde (cinnamon), eugenol (incl. clove), thymol (incl. thyme) and menthol (incl. mint) exhibited a similar but weaker effect.

Transient Receptor Potential (TRP) channels are cell parts that have been preserved during evolution. They constitute the biological sensors which perceive ambient changes as a response to stimuli such as heat or cold, mechanical forces and natural chemical substances. TRP channels are ion channels which are permeable to cations; they are Ca 2+ permeable and are of great importance in the sensory transduction of visual light and sound stimuli, pheromone effects, taste and temperature sensations, pain perception, perception of corrosive excitatory substances, renal Ca2+/Mg2+ regulation, smooth muscle tone, blood pressure regulation and maintaining cell redox potential. Modulation of these channels by on the one hand retarding and on the other activating different types of TRP channels, which carvacrol is able to do, results in reduced cell stress, which in vitro leads to a higher survival rate of hippocampal neurones under anoxic conditions (ischaemia). More specifically, carvacrol inhibits the TRPM-7 channel (TRPMelastatin7) which shows increased activity during ischaemia (hypoxia). Synthetic anti-excitatory remedies and NMDA receptor antagonists have proven to be of little clinical value until now in preventing ischaemic damage to the brain. Researchers suggest that carvacrol is a potential candidate for limiting brain damage as a result of ischaemia. 

Carvacrol and thymol exhibit a blood vessel relaxant effect on the aorta tissue in rats. This effect is not dependent on the endothelial cells and possibly occurs via the modulation of Ca2+ release by the sarcoplasmatic reticulum on the one hand and/or by controlling the susceptibility to Ca2+ of the contractile system of the blood vessel on the other. It is plausible that carvacrol in low concentrations inhibits the influx of Ca2+ and thus relaxes the smooth muscle tissue of the blood vessel. 

Infection inhibiting and anti-oxidative properties

Oral administration of water-ethanol extract of wild oregano in mice significantly prevents the occurrence of gastritis within the cold-restraint stress model. Higher doses of 100 mg/kg and 200 mg/kg of bodyweight hardly resulted in any additional therapeutic benefit compared with the lowest used dosage of 30 mg/kg. 

There was a clear correlation between the gastritis caused by stress and the degree of oxidative stress in laboratory animals. The anti-oxidative properties of wild oregano oil contribute to its preventive characteristics to a significant degree.

The anti-oxidative substances in wild oregano oil such as rosemary acid, coffee acid and various flavonoids therefore function directly as anti-oxidants as well as ion donors to tissue-protecting peroxidases (POD) in saliva (sPOD) and the mucous membrane (mPOD) in the alimentary canal.

Percutaneous administration of oregano oil also prevented contact hyper-susceptibility to the test substance oxazolone, often used for this purpose. The infection inhibiting properties are nevertheless less effective than those of hydrocortisone.

Cyclooxygenase-2 (COX-2) is the dominant enzyme in determining the biosynthesis of prostaglandins. It plays a key role in infections and in circulatory homeostasis including glucose and fat metabolism. COX-2 expression is controlled by ligand-dependent transcription factors such as PPARs (Peroxisomal Proliferator-Activated Receptors) which belong to a group of nuclear cell receptors. PPARs control the COX-2 expression and vice versa. Carvacrol turns out to be a powerful suppressor of the COX-2 expression and an activator of PPAR-alpha and -gamma. These characteristics of carvacrol are representative of its infection inhibiting and lifestyle compensatory effects and may potentially reduce the metabolism-disrupting effects caused by stress on humans. 

In rats, carvacrol strongly reduces the toxic effect of D-galactosamine on the liver. As a result of D-galactosamine, the asat (aspartate-amino-transferase), alat (alanine aminotransferase) and ggt (gamma glutamyltranspeptidase) levels in VLDL and LSL cholesterol and liver enzymes increase. The HDL cholesterol is lowered at the same time while the total cholesterol, triglycerides and free fatty acids increase. However, if D-galactosamine is combined with carvacrol for a period of 21 days, all these parameters improve towards normal levels.

It is interesting to note that in order to generate lethal hepatitis for research purposes, D-galactosamine is combined with LPS (lipopolysaccharide of a pathogenic type), which is a constituent of the cell wall of pathogenic intestinal bacterial. D-galactosamine increases the susceptibility of the liver to LPS, which releases large quantities of TNF-alpha causing lethal hepatitis. 

Anticipating the following description of the properties of carvacrol-rich oregano oil, this oil could, in the case of a highly permeable intestine, also potentially decrease the susceptibility of the liver and the portal vein system as regards LPS, which would modulate the inflammatory response as a result. As can be further deduced from the properties of oregano oil, this throws a hypothetical light on the therapeutic approach to many western health issues such as metabolic syndrome, heart and vascular disease and type 2 diabetes.  

Metabolism of glucose and fat 

Oregano oil and its constituent substances, such as gamma-terpinene and carvacrol, have demonstrated in various in vitro studies that they have the ability to inhibit the aggregation and adhesion of blood platelets and cholesterol synthesis, whereas a decline was observed in vivo in total cholesterol, triglycerides and both the systolic and diastolic readings in hypertension. 

DNA protection

Carvacrol was added to the drinking water of rats for a period of 7 or 14 days during a research study, which resulted in reduced DNA damage in both groups compared to the control group which drank untreated tap water. The lowest dosage was 15 mg of carvacrol per kilogram per day. The isolated liver and testis cells of animals that were given carvacrol, turned out to be less susceptible to DNA damage when they were exposed to hydroperoxide. The researchers assume that carvacrol causes the anti-oxidative capacity of the liver and testes cells to increase.

Antibacterial and antimycotic properties

Research into preventing food decay and growth of pathogens in particular has shown that carvacrol is effective in killing or decimating bacteria, yeasts, fungi, insects and mites. In one pilot in vitro study, carvacrol also killed the corona virus, which is often involved in colds and influenza.

The bactericidal effect of carvacrol is based on enhanced proton and ion permeability in the cell membrane, which makes the cell lose its integrity so the essential cell elements such as nucleid acid and ATP leech away. It has in addition been shown that in the case of Escherichia coli, carvacrol inhibits the ATPase enzyme and induces the heat shock proteins Hsp60 and Hsp70. This results in a decrease in motility which inhibits the development of flagella. 

The anti-microbial effect has been demonstrated in vitro against dozens of micro-organisms and has again recently been confirmed through in vitro research for four Gram positive bacteria.

 Staphyllococcus aureus, S. epidermidis, S. mutans and S. viridans, four Gram negative bacteria: Penicillium aeruginosa, Escherichia coli, E. cloacae and Klebsiella pneumonia as well as against three pathogenic fungi: Candida albicans, C. tropicalis and C. glabrata. 

Both in vitro and in vivo research shows that wild oregano oil has a curative effect on mice that have been injected with Staphylococcus aureus. Untreated mice died within a week, but when oregano oil was administered, one-third of the animals survived. Treatment with monolaurin from coconut fat resulted in 50% survival, which is at a level equal to that of animals treated with the antibiotic Vancomycin. The combination of wild oregano oil with monolaurin that is created in the body from lauric acid (coconut oil) exhibits a superior effect with a survival rate of over 60%. In humans, wild oregano oil and/or monolaurin could act both preventively and curatively against Staphylococcus aureus infections, because resistant MRSA bacteria are also susceptible to wild oregano oil.

In vitro research shows that a natural mix of phenols from oregano has an anti-bacterial effect on Helicobacter pylori, which is held to be responsible for many types of stomach complaints and infections. 

In vitro, the fungistatic and fungicidal effect of wild oregano oil is comparable to that of synthetic carvacrol, olive oil, amphotericin B and nystatin. The latter two were effective in lower concentrations. However, the minimum concentration required to kill Candida albicans in vitro was twice as high for pure carvacrol as it was for wild oregano oil.

In vivo, the oral administration of wild oregano oil is as effective as synthetic carvacrol in fighting systemic candidiasis in mice. Mice infected with five times the lethal dose of candida albicans died within 10 days if they were not treated with carvacrol or wild oregano oil. A daily dose of wild oregano oil of 17.33 mg/kg of bodyweight resulted in 100% survival after 30 days. Cosmetically, the mice which were treated with the natural wild oregano oil looked the best. In vitro research shows that even low concentrations of wild oregano oil will inhibit growth of pseudohyphae or germ tubes. By way of explanation, the growth of germ tubes is generally considered to be the phenotypic plasticity whereby Candida starts to exhibit invasive characteristics. 

Inhibition of elastase

It appears that wild oregano oil, and carvacrol in particular, inhibit the activity of HLE (Human Neutrophil Elastase) when used in low doses, which is why wild oregano oil could potentially be a phytotherapeutic in diseases such as COPD and emphysema.

Inhibiting acetylcholinesterase

In vitro, carvacrol exhibits a cholinesterase inhibiting effect that is ten times stronger than that of its thymol isomer. The cholinesterase inhibiting property may be useful in combating cognitive deterioration as found, for example, in dementia.

Anxiolytic and antidepressant properties

Tests and research into the behaviour of mice show an anxiolytic effect as regards one-off administration of carvacrol.  The effects of both carvacrol (12.5; 25 and 50 mg/kg) and diazepam (1 and 2 mg/kg) were cancelled out by flumazenil (benzodiazepine antagonist).

Carvacrol administered in the same dosage resulted in an antidepressant effect when mice were exposed to stressful tests. The effect was attributed to an interaction with the dopaminergic system but not to the serotonergic or the noradrenergic systems. 

Intestinal cleansing effect

Piglets which are given feed mixed with carvacrol or thymol after weaning, exhibit a higher villus to crypt ratio in the entire small intestine than the control group which was not given the supplement. This was demonstrated by the use of several experimental feeds. The experimental fodder contained 500 and 2000 mg carvacrol or thymol per kilogram of feed.  In this research programme, carvacrol and thymol improved the health of the intestine, which was confirmed by a histological investigation. However, in no part of the intestines was the number of bacteria reduced by carvacrol or thymol. 

As a result of the ingestion of food with 2000 mg carvacrol per kilogram of feed, the carvacrol concentration in the stomach and the top of the small intestine was respectively 521 and 5 mg per kilogram of chyme. The reabsorption of carvacrol therefore appears to be more than 90%.  

Anti-parasitic 

An emulsion of wild oregano oil at 600 mg per day for 6 weeks given to patients with intestinal parasites (Blastocystis hominis, Entamoebe hartmanni and Endolimax nana) resulted in complete elimination of Entamoebe hartmanni (4 patients), Endolimax nana (1 patient) and Blastocystis hominis (8 patients). The Blastocystis hominis score diminished in 3 other patients. In 7 of these 11 patients with Blastocystis hominis the stomach and intestinal symptoms improved.

In vitro research further shows that carvacrol-rich wild oregano oil can be clinically useful in the treatment of Giardia lamblia. The concentration of carvacrol-rich wild oregano oil of two different kinds ranged from 71 to 85 microgram/ml and blocked the adhesive characteristics of the parasite, whereby during the following period half of the number of parasites were also killed. 

A single in vitro study which compared wild oregano oil with Tinidazole, an existing anti-giardia remedy, even showed that wild oregano oil was more effective.

INDICATIONS

On contemplating wild oregano oil and the effects and mechanisms observed so far, it would be possible to represent the therapeutic bandwidth of wild oregano oil among other things by the inhibiting effect on provocations by bacterial LPS from the human intestine via the portal vein system in the liver. Such provocations via the human intestine form part of an explanatory model of the persistence of a low-grade infection syndrome as a causal mechanism for many pathologies such as metabolic syndrome, heart and vascular disease, type 2 diabetes and various ageing related (neuro-)degenerative diseases. If we also take the observations on board which indicate an improvement in health of the small intestine and the villi, as well as the property of decimating opportunists such as Candida and other intestinal pathogens, one cannot deny that wild oregano oil has great clinical potential. Moreover, its specific infection inhibiting and PPAR-gamma activating features contribute to the reduction of insulin resistance and the normalisation of glucose and fat metabolism. Additional in vivo studies and clinical research into the use of wild oregano oil in humans will be needed in order to determine its further medical significance. 

Candidiasis and parasitic infections

Wild oregano oil has so far been prescribed by many therapists for treatment of gastrointestinal candidiasis (GIC). This kind of application is based on clinical experience. For example, in cases of patients with chronic fatigue syndrome, an oral caprylic acid preparation was prescribed as an initial treatment for GIC. Administration was then changed to oral administration of a vegetable oil such as wild oregano oil, to fight excess Candida growth in skin folds, nails, ears and sinuses. 

In vitro research shows that caprylic acid in high doses works quickest against Candida, but that lauric acid in low doses is superior. The minimal growth-inhibiting concentration against Candida is however the lowest for oregano oil, while a convincing systemic anti-Candida effect has only been demonstrated in vivo in mice for wild oregano oil.

Besides this, wild oregano oil could be of use in the treatment of a number of intestinal parasites such as Blastocystis hominis, Entamoebe hartmanni and Endolimax nana.

In vitro research also shows that wild oregano oil has anti-parasitic properties against Giardia lamblia. The research into the anti-parasitic effect of wild oregano oil is rather meagre so far. In short, in line with traditional uses, wild oregano oil appears to show its value in interventions for cleansing the alimentary and immune systems. Furthermore, wild oregano oil is traditionally used against mental exhaustion and has a calming and strengthening effect.

SIDE EFFECTS

Oregano and wild oregano oil are considered a safe food additive in America and Europe. Moreover, no undesirable side effects from wild oregano oil are known. 

Wild oregano oil with carvacrol as the major constituent proves to be non-toxic in acute and chronic pre-clinical toxicity models. Carvacrol (5-isopropyl-2-methylphenol) is not hepatotoxic to rats. Research into separate oil fractions of oregano showed that carvacrol, in contrast to thymol, is not an irritant to the skin. There are a few known cases of a systemic allergic reaction as the result of ingestion of oregano. 

The pure essential oil of wild oregano can also cause irritation of the mucous membranes. That is why a concentration of 1% is recommended for direct application to the mucous membranes.  

Given that the internal as well as the oral therapeutic application of wild oregano oil in humans has hardly been researched, it is not recommended to use the oil in pregnancy. The German E-Commission does not publish any advice on therapeutic use because it feels that the effects have been insufficiently documented.

SYNERGISM

Lauric acid from coconut oil breaks down in the body into monolaurin, which in vitro exhibits a similar effect to that of wild oregano oil on micro-organisms and works synergetically against Staphylococcus aureus, as demonstrated in vivo. In vitro research also shows an anti-bacterial synergy as regards phenols from oregano and cranberries against Helicobacter pylori.

There is in addition a potential synergy with the use of probiotics, because clinical studies and in vivo research into rats show that probiotics (Lactobacillus acidophilus) appear to be effective against fungal mucosal colonisations by Candida glabrata and Candida albicans in the digestive tract. In these cases, probiotics demonstrably help to heal ulcerations and infections. It has also been shown that Lactobacillus acidophilus reduces the duration of a fungal colonisation.

Origanum vulgare induces apoptosis in human colon cancer caco2 cells.

Oregano spice is widely used in the Mediterranean diet, which is associated with a low risk for colon cancer. Although the medicinal benefits of oregano, such as the anti-inflammatory and antimicrobial activities, are well known; nonetheless, only few data are available on its effect in cancer prevention, especially concerning the mechanism of action. Here, we investigated the effect of Origanum vulgare ethanolic extracts on redox balance, cell proliferation, and cell death in colon adenocarcinoma Caco2 cells. Oregano extract leads to growth arrest and cell death in a dose- and time-dependent manner. Changes in glutathione content, as well as the increase in its oxidized form, may be involved in oregano-triggered death. Both extrinsic and intrinsic apoptotic pathways appear to be activated by spice extract. Our findings suggest that oregano amounts found in the Mediterranean diet can exert proapoptotic effects, which are selective for cancer cells. Moreover, whole extract, instead of a specific component, can be responsible for the observed cytotoxic effects.

The effect of an aqueous extract of Origanum vulgare (OV) leaves on blood glucose levels was investigated in normal and streptozotocin (STZ) diabetic rats. In normal rats, the blood glucose levels were slightly decreased 6 h after a single oral administration (P<0.05) as well as 15 days after once daily repeated oral administration of aqueous OV extract (P<0.05) (20 mg/kg). After a single dose or 15 daily doses, oral administration of the aqueous extract (20 mg/kg) produced a significant decrease on blood glucose levels in STZ diabetic rats (P<0.001). In STZ rats, the blood glucose levels were normalised from the fourth day after daily repeated oral administration of aqueous OV extract (20 mg/kg) (P<0.001). However, this effect was less pronounced 2 weeks after daily repeated oral administration of OV extract. In addition, no changes were observed in basal plasma insulin concentrations after treatment in either normal or STZ diabetic rats indicating that the aqueous OV extract acted without changing insulin secretion. We conclude that an aqueous extract of OV exhibits an anti-hyperglycaemic activity in STZ rats without affecting basal plasma insulin concentrations.

The present investigation is focused on antibacterial potential of infusion, decoction and essential oil of oregano (Origanum vulgare) against 111 Gram-positive bacterial isolates belonging to 23 different species related to 3 genera. Infusion and essential oil exhibited antibacterial activity against Staphylococcus saprophyticus, S. aureus, Micrococcus roseus, M. kristinae, M. nishinomiyaensis, M. lylae, M. luteus, M. sedentarius, M. varians, Bacillus megaterium, B. thuringiensis, B. alvei, B. circulans, B. brevis, B. coagulans, B. pumilus, B. laterosporus, B. polymyxa, B. macerans, B. subtilis, B. firmus, B. cereus and B. lichiniformis. The infusion exhibited maximum activity against B. laterosporus (17.5 mm mean zone of inhibition+/-1.5 Standard deviation) followed by B. polymyxa (17.0 mm+/-2.0 SD) and essential oil of oregano exhibited maximum activity against S. saprophyticus (16.8 mm+/-1.8 SD) followed by B. circulans (14.5 mm+/-0.5 SD). While all these tested isolates were found resistant to decoction of oregano.

The aim of this study was to evaluate the in vitro activity of the essential oil extracted from Origanum vulgare against sixteen Candida species isolates. Standard strains tested comprised C. albicans (ATCC strains 44858, 4053, 18804 and 3691), C. parapsilosis (ATCC 22019), C. krusei (ATCC 34135), C. lusitaniae (ATCC 34449) and C. dubliniensis (ATCC MY646). Six Candida albicans isolates from the vaginal mucous membrane of female dogs, one isolate from the cutaneous tegument of a dog and one isolate of a capuchin monkey were tested in parallel. A broth microdilution technique (CLSI) was used, and the inoculum concentration was adjusted to 5 x 106 CFU mL-1. The essential oil was obtained by hydrodistillation in a Clevenger apparatus and analyzed by gas chromatography. Susceptibility was expressed as Minimal Inhibitory Concentration (MIC) and Minimal Fungicidal Concentration (MFC). All isolates tested in vitro were sensitive to O. vulgare essential oil. The chromatographic analysis revealed that the main compounds present in the essential oil were 4-terpineol (47.95%), carvacrol (9.42%), thymol (8.42%) and □-terpineol (7.57%). C. albicans isolates obtained from animal mucous membranes exhibited MIC and MFC values of 2.72 μL mL-1 and 5 μL mL-1, respectively. MIC and MFC values for C. albicans standard strains were 2.97 μL mL-1 and 3.54 μL mL-1, respectively. The MIC and MFC for non-albicans species were 2.10 μL mL-1 and 2.97 μL mL-1, respectively. The antifungal activity of O. vulgare essential oil against Candida spp. observed in vitro suggests its administration may represent an alternative treatment for candidiasis

In this study, the antioxidative capacity effect of essential oils and aqueous tea infusions obtained from oregano, thyme and wild thyme on the oxidation susceptibility of low-density lipoproteins (LDL) has been studied. The results indicate a dose-dependent protective effect of the tested essential oils and aqueous tea infusions on the copper-induced LDL oxidation. The protective effect of essential oils is assigned to the presence of phenolic monoterpenes, thymol and carvacrol, which are identified as the dominant compounds in these essential oils. The strong protective effect of aqueous tea infusions is proposed to be the consequence of large amounts of polyphenols, namely rosmarinic acid and flavonoids (quercetin, eriocitrin, luteolin-7-O-glucoside, apigenin-7-O-glucoside, luteolin, apigenin), with the most pronounced effect in the case of oregano. These findings may have implications for the effect of these compounds on LDL in vivo.

This research was aimed at investigating the essential oil production, chemical composition and biological activity of a crop of pink flowered oregano (Origanum vulgare L. subsp. vulgare L.) under different spatial distribution of the plants (single and binate rows). This plant factor was shown to affect its growth, soil covering, fresh biomass, essential oil amount and composition. In particular, the essential oil percentage was higher for the binate row treatment at the full bloom. The chemical composition of the oils obtained by hydrodistillation was fully characterized by GC and GC-MS. The oil from plants grown in single rows was rich in sabinene, while plants grown in double rows were richer in ocimenes. The essential oils showed antimicrobial action, mainly against Gram-positive pathogens and particularly Bacillus cereus and B. subtilis.

This investigation focused on the qualitative and quantitative composition of essential oil compounds of European Origanum vulgare. Extracts of 502 individual O. vulgare plants from 17 countries and 51 populations were analyzed via GC. Extracts of 49 plants of 5 p opulations of Israeli Origanum syriacum and 30 plants from 3 populations of Turkish Origanum onites were included to exemplify essential oil characteristics of 'high-quality' oregano. The content of essential oil compounds of European O. vulgare ranged between 0.03% and 4.6%. The monoterpenes were primarily made up of sabinene, myrcene, p-cymene, 1,8-cineole, β-ocimene, γ-terpinene, sabinene hydrate, linalool, α-terpineol, carvacrol methyl ether, linalyl acetate, thymol and carvacrol. Among the sesquiterpenes β-caryophyllene, germacrene D, germacrene D-4-ol, spathulenol, caryophyllene oxide and oplopanone were often present in higher amounts. According to the proportions of cymyl-compounds, sabinyl-compounds and the acyclic linalool/linalyl acetate three different main monoterpene chemotypes were defined. The cymyl- and the acyclic pathway were usually active in plants from the Mediterranean climate whereas an active sabinyl-pathway was a characteristic of plants from the Continental climate.


Effect of Pretreatment With Extract of Origanum vulgare Leaves on Experimental Intestinal Ischemia-Reperfusion Injury in Rats Omid Azari,1,* Reza Kheirandish,2 Hamideh Rohani,3 and Saeedeh Shojaeepour4

Background: Origanum vulgare is used in traditional medicine for antimicrobial, anti-inflammatory and antioxidant activities.

Objectives: The present study was designed to investigate the protective effects of O. vulgare leaves extract on experimentally induced intestinal ischemia/reperfusion (I/R) injury in rats.

Materials and Methods: In this experimental study, 24 male Wistar albino rats randomly divided into four groups. Group I/R underwentischemia-reperfusion of the intestine (45 minutes of ischemia followed by 1 hour of reperfusion). Treatment groups I and IIwere givenO. vulgare extract (200 and 400 ppm) via oral gavages for 1 week before inducing I/R. Group sham was given normal saline orally without inducing I/R. After the experiments, thejejunum was removed and the tissues were processed for histomorphometric examination of mucosa.

Results: The treatment with O. vulgare extract significantly decreased mucosal damages in the treatment groups compared to groupI/R, while severe mucosal damages were observed in the group I/R. Also, there was significant difference between treatment groupsI and II (P = 0.054). Group sham observed normal intestinal mucosa.

Conclusions: According to the results of current study, administration of O. vulgare extract protects the intestinal mucosa from I/R injuries.


Methanolic extract of Origanum vulgare ameliorates type 1 diabetes through antioxidant, anti-inflammatory and anti-apoptotic activity Milica Vujicica1, Ivana Nikolica1, Vassiliki G. Type 1 diabetes (T1D), an autoimmune inflammatory disorder, develops as a consequence of pancreatic β-cell destruction and results in hyperglycaemia. Since current T1D therapy mainly involves insulin replacement, the aim of the present study was to evaluate the therapeutic potential of Origanum vulgare L. ssp. hirtum (Greek oregano) leaf extract rich in biophenols for the treatment of T1D. The phytochemical profile of methanolic oregano extract (MOE) and aqueous oregano extract (AOE) was determined by liquid chromatography/electrospray ion-trap tandem MS (LC/DAD/ESI-MSn), while their main compounds were quantified by HPLC with diode array detection. After establishing their potent in vitro antioxidant activity, the extracts were administered to C57BL/6 mice treated with multiple low doses of streptozotocin for diabetes induction. While prophylactic AOE therapy had no impact on diabetes induction, MOE reduced diabetes incidence and preserved normal insulin secretion. In addition, MOE scavenged reactive oxygen and nitrogen species and, therefore, alleviated the need for the up-regulation of antioxidant enzymes. MOE treatment specifically attenuated the pro-inflammatory response mediated by T helper 17 cells and enhanced anti-inflammatory T helper 2 and T regulatory cells through the impact on specific signalling pathways and transcription factors. Importantly, MOE preserved β-cells from in vitro apoptosis via blockade of caspase 3. Finally, rosmarinic acid, a predominant compound in MOE, exhibited only partial protection from diabetes induction. In conclusion, acting as an antioxidant, immunomodulator and in an anti-apoptotic manner, MOE protected mice from diabetes development. Seemingly, there is more than one compound responsible for the beneficial effect of MOE.

Wilde marjolein / Origanum vulgare monografie

Wilde oregano (Origanum vulgare) behoort tot de groep van Lamiaceae waartoe ook rozemarijn, tijm en munt behoren. Origanum is de klassieke Griekse naam welke is afgeleid van de woorden ‘Oros’ (‘Berg’) en ‘Ganos’ (‘Vreugde’) die verwijzen naar de mooie verschijning in de bergen, ofwel van ‘Origanon’ dat ‘bitter kruid’ betekent. 

Wilde oregano (Origanum vulgare), verwarrend genoeg ook wel “wilde marjolein” genoemd, is een kruid dat rijk is aan het fenol carvacrol dat  zich kenmerkt door een pittige bittere smaak met een lichte warmtesensatie. Dit in tegenstelling tot Origanum majorana of echte marjolein dat ook wel zoete oregano wordt genoemd en veel in de keuken wordt gebruikt, maar minder rijk is aan therapeutische substanties. Ter onderscheiding van zoete oregano c.q. echte marjolein wordt in deze tekst voor de bittere (en carvacrolrijke) Origanum vulgare de term “wilde oregano” gebruikt.

De meest krachtige wilde origanumvariëteiten groeien in de oostelijke mediterrane berggebieden op een kalkrijke grond. Carvacrol is de meest onderzochte inhoudsstof en synthetisch carvacrol wordt al meer dan vijftig jaar gebruikt in reinigingsmiddelen als lysol en Listerine om instrumentaria en werkoppervlakken te desinfecteren. Heden ten dage is de interesse in de oorspronkelijke plantensubstanties weer actueel, omdat de resistentie van bacteriën en schimmels tegen moderne medicijnen als antibiotica ons nopen nieuwe oplossingen te vinden. Vele plantaardige substanties blijken in meer of mindere mate een breedspectrum therapeuticum te zijn tegen zowel bacteriën, gisten, schimmels, parasieten als virussen. Wilde oregano-olie lijkt in dit opzicht een alleskunner. Veterinair onderzoek ten behoeve van de veeteelt en pluimveehouderij ziet mogelijkheden om het antibioticagebruik terug te dringen middels het gebruik van wilde oregano-olie in voeders. De humane voedingsindustrie is meer geïnteresseerd in wilde oregano-olie als natuurlijk conserveermiddel. De oude Egyptenaren gebruikten oregano al om voeding te conserveren en wonden te reinigen. De oude Grieken pasten het ook toe bij hoofdpijn, zweren, longziekten, astma en spasmen. In het huidige Griekenland wordt thee van oregano als volksmedicijn gebruikt bij verkoudheden, keelpijn, griep, maagzweren en andere maag-darmklachten.

De effectiviteit van de carvacrolrijke etherische olie van wilde oregano blijkt qua veelzijdigheid en effectiviteit te excelleren. De effecten zijn dosisafhankelijk en tot dusver wordt iedere microbe gedood of in zijn groei geremd wanneer het wordt blootgesteld aan de etherische olie van de wilde oregano. Oregano-olie vertoont een breed anti-microbieel effect tegen voedselpathogenen en bevat potente antioxidanten die in onderzoek een therapeutische waarde vertegenwoordigen voor het cardiovasculaire systeem en het zenuwstelsel. Oregano werkt ontstekingsremmend en reguleert het glucose- en vetmetabolisme. Ondanks het toenemend aantal in-vivostudies ontbreekt het nog aan goed placebogecontroleerd klinisch onderzoek bij mensen. De rijke historie en de positieve anekdotes zijn mede representatief voor de potentiële klinische waarde van wilde oregano-olie.

Eigenschappen:

Inhoudstoffen

De etherische olie van de wilde oreganovariëteiten bestaat vooral uit carvacrol gevolgd door zijn precursors p-cymeen en gammaterpineen. Dit zijn ook de precursors voor thymol dat een isomeer is van carvacrol maar dat in wilde oregano-olie uit de oostelijke mediterrane gebergten slechts een klein percentage vormt ten opzichte van carvacrol. Van nature kunnen concentraties verschillen. Halffabricaten van de etherische olie kunnen worden gestandaardiseerd voor verdere verwerking.

Werkingsmechanismen:

In-vitro-onderzoek toont dat carvacrol tot dusver de meest potente modulator is van de TRP-kanalen in de cellen van zoogdieren. Andere substanties als kaneelaldehyde (cinnamaldehyde), eugenol (oa. kruidnagel), thymol (o.a. tijm) en menthol (o.a. munt) vertoonden een gelijkaardig effect dat echter zwakker was. Transient Receptor Potential (TRP) kanalen zijn evolutionair geconserveerde celonderdelen. Ze vormen biologische sensoren die omgevingsveranderingen kunnen waarnemen als respons op stimuli als warmte of koude, mechanische krachten en natuurlijke chemische substanties. TRP-kanalen zijn kationpermeabele ionkanalen, ze zijn Ca 2+ doorlaatbaar en van groot belang bij de sensorische transductie van visuele lichtprikkels, geluid, feromooneffecten, smaakperceptie, temperatuursensatie, pijnperceptie, perceptie van bijtende prikkelende substanties, renale Ca2+/Mg2+ regulatie, tonus van glad spierweefsel, bloeddrukregulatie en handhaving van de redoxstaat van een cel. Modulatie van deze kanalen via enerzijds remmen en anderzijds activeren van verschillende type TRP-kanalen zoals carvacrol dat kan, resulteert in het verlagen van celstress hetgeen in vitro bij anoxie (ischemie) leidt tot een hogere overleving van hippocampale neuronen. Meer specifiek remt carvacrol het TRPM7-kanaal (TRPMelastatin7) dat verhoogd actief wordt bij ischemie (hypoxie). Tot dusver blijken synthetische anti-excitatoire medicijnen en NMDA-receptorantagonisten klinisch van weinig waarde in het voorkomen van ischemische schade aan het brein. De onderzoekers suggereren dat carvacrol een potentiële kandidaat is voor het beperken van hersenschade als gevolg van ischemie. 

Carvacrol en thymol tonen een bloedvatrelaxerend effect op aortaweefsel van ratten. Dit effect is niet endotheelafhankelijk en geschiedt mogelijk via het moduleren van enerzijds  Ca 2+ vrijzetting door het sarcoplasmatisch reticulum en/of anderzijds door regulatie van de Ca2+ gevoeligheid van het contractiele systeem van het bloedvat. Aannemelijk is dat carvacrol in lage concentraties de Ca2+ influx remt en zo het gladde spierweefsel van het bloedvat ontspant. 

Orale toediening van water-ethanolextract van wilde oregano bij muizen voorkomt significant het ontstaan van gastritis binnen het “cold-restraint stress model”. Hogere doseringen van 100 en 200 mg/kg lichaamsgewicht leverden in vergelijking met de laagst gebruikte dosering van 30 mg/kg maar nauwelijks extra therapeutisch voordeel op. Er was bij de proefdieren een duidelijk verband tussen de door stress veroorzaakte gastritis en de mate van oxidatieve stress. De antioxidatieve eigenschappen van wilde oregano-olie dragen in sterke mate bij aan de preventieve eigenschappen. De anti-oxidatieve substanties in wilde oregano-olie zoals rozemarijnzuur, koffiezuur en diverse flavonoïden fungeren behalve direct als antioxidant tevens als iondonor voor weefselbeschermende peroxidases (POD) in het speeksel (sPOD) en de mucosa (mPOD) van het spijsverteringskanaal. Percutane toediening van oregano-olie voorkwam tevens contactovergevoeligheid voor de daarvoor vaak gebruikte teststof oxazolon. De ontstekingsremmende eigenschappen zijn echter minder dan die van hydrocortisol.

Cyclooxygenase-2 (COX-2), is het meest bepalende enzym in de biosynthese van prostaglandines. Het speelt een sleutelrol bij ontstekingen en de circulatoire homeostase waaronder het glucose- en vetmetabolisme. COX-2-expressie wordt gereguleerd door ligandafhankelijke trascriptiefactoren als de PPARs (Peroxisome Proliferator-Activated Receptors) welke behoren tot groep van celkernreceptoren. De PPARs reguleren de COX-2-expressie en vice versa. Carvacrol blijkt een krachtige suppressor te zijn van de COX-2-expressie en een activator van PPAR-alfa en -gamma. Deze eigenschappen van carvacrol zijn representatief voor de ontstekingsremmende en leefstijlcompenserende effecten en zouden in potentie het metabolisme-ontregelende effect van stress op de mens kunnen verminderen. 

Carvacrol vermindert bij ratten sterk het toxische effect van D-galactosamine op de lever. Als gevolg van D-galactosamine stijgt het VLDL- en LDL-cholesterol en de leverenzymen asat (aspartaat-amino-transferase), alat (Alanine aminotransferase) en ggt (gamma glutamyltranspeptidase). Daarbij daalt het HDL-cholesterol en neemt in de lever en nieren het totaalcholesterol, trigyceriden en vrije vetzuren toe. Indien echter D-galactosamine gedurende 21 dagen werd gecombineerd met carvacrol verbeterde al deze parameters zich richting normaalwaarden. Interessant is dat voor het opwekken van een letale hepatitis ten behoeve van onderzoek, D-galactosamine wordt gecombineerd met LPS (lipopylsacharide van pathogene aard), een celwandcomponent van pathogene darmbacteriën. D-galactosamine verhoogt de gevoeligheid van de lever voor LPS waardoor in hoge mate TNF-alfa wordt vrijgezet en een letale hepatitis ontstaat. Vooruitlopend op de verder beschreven eigenschappen van carvacrolrijke oregano-olie zou deze olie mogelijk ook bij een verhoogd permeabele darm de gevoeligheid van de lever en het poortadersysteem voor LPS uit de darminhoud kunnen verminderen waardoor de inflammatoire respons gemoduleerd wordt. Zoals verder uit de eigenschappen van oregano-olie op te maken valt,  werpt dit een hypothetisch licht op de therapeutische benadering van veel westerse gezondheidsproblemen als het metabool syndroom, hart- en vaatziekten en diabetes type 2.

Oregano-olie en inhoudstoffen zoals gamma-terpineen en carvacrol hebben in verscheidene in vitro studies aangetoond dat ze de aggregatie en adhesie van bloedplaatjes en de cholesterolsynthese kunnen remmen waarbij in vivo een daling van het totaalcholesterol, triglyceriden en de bloeddruk (systolisch en diastolisch) werd waargenomen. 

In een onderzoek waarbij gedurende 7 of 14 dagen carvacrol aan het drinkwater van ratten werd toegediend, resulteerde dit voor beide groepen in minder DNA-schade in vergelijking met de controlegroep met onbehandeld kraanwater. De laagste dosering was 15 mg carvacrol per kilogram per dag. De geïsoleerde lever- en testiscellen van dieren die carvacrol kregen bleken minder gevoelig voor DNA-schade wanneer ze werden blootgesteld aan waterstofperoxide. De onderzoekers veronderstellen dat carvacrol de antioxidatieve capaciteit van lever en testiscellen doet toenemen.

Vooral onderzoek naar het voorkomen van voedselbederf en groei van pathogenen heeft aangetoond dat carvacrol effectief is in het afdoden of decimeren van bacteriën, gisten, schimmels, insecten en mijten. In een in-vitropilotstudie bleek carvacrol ook het coronavirus te doden dat veelal betrokken is bij verkoudheden en griep.

Het bacteriedodend effect van carvacrol berust op het verhoogd doorlaatbaar maken van het celmembraan voor protonen en ionen, waardoor het zijn integriteit verliest en essentiële celcomponenten als nucleïnezuren en ATP weglekken. Daarbij is bij Escherichia coli aangetoond dat carvacrol het enzym ATPase remt en Hsp60 en Hsp70 (heat shock protein) induceert. Dit resulteert in een afname van de motiliteit waarbij tevens de ontwikkeling van flagella (zweepstaartjes) wordt geremd. Het antimicrobiële effect is in vitro al tegen vele tientallen micro-organismen aangetoond en werd recentelijk via in-vitro-onderzoek nogmaals bevestigd voor vier grampositieve bacteriën: Stafyllococcus aureus, S. epidermidis, S. mutans en S. viridans, vier gram-negatieve bacteriën: Penicillium aeruginosa, Escherichia coli, E. cloacae en Klebsiella pneumonia alsmede tegen drie pathogene schimmels: Candida albicans, C. tropicalis en C. glabrata. 

In-vitro- en in-vivo-onderzoek toont dat wilde oregano olie curatief werkt wanneer muizen worden geïnjecteerd met Stafylococcus aureus. Onbehandelde muizen stierven binnen een week, maar door toediening van oregano-olie overleefde een derde van de dieren. Behandeling met monolaurine uit kokosvet resulteerde in 50% overleving hetgeen gelijk is aan met Vancomycine (antibiotica) behandelde dieren. De combinatie van wilde oregano olie met monolaurine dat in het lichaam ontstaat uit laurinezuur (kokosolie) vertoont met meer dan 60% overleving een superieur effect. Wilde oregano-olie en/of monolaurine zouden bij de mens zowel preventief als curatief kunnen dienen tegen infecties met Stafylococcus aureus, omdat ook de resistente MRSA-bacterie gevoelig is voor wilde oregano-olie. In-vitro-onderzoek toont dat een natuurlijke mix van fenolen uit oregano een antibacterieel effect heeft op de Helicobacter pylori die verantwoordelijk wordt gehouden voor veel soorten maagklachten en -ontstekingen. 

In vitro is het fungistatische en fungicide effect van wilde oregano-olie vergeleken met synthetisch carvacrol, olijfolie, amphotericine B en nystatine. De laatste twee waren in lagere concentraties effectief. De minimale concentratie om Candida albicans in vitro te doden was voor puur carvacrol echter twee maal zo hoog als voor wilde oregano-olie. In vivo is de orale toepassing van oregano-olie even effectief als synthetisch carvacrol in het bestrijden van een systemische candidiasis bij muizen. Muizen geïnfecteerd met vijf maal de letale dosis candida albicans stierven binnen 10 dagen indien ze niet behandeld werden met carvacrol of oregano-olie. Een dagelijkse dosering wilde oregano-olie van 17,33 mg/kg lichaamsgewicht resulteerde na 30 dagen in een 100% overleving. De muizen die behandeld werden met de natuurlijke oregano-olie zagen er na behandeling cosmetisch het beste uit. In vitro onderzoek toont dat zelfs zeer lage concentraties wilde oregano-olie al de vorming van kiembuisjes (pseudohyfe of germtubes) remt. Ter verduidelijking, de vorming van kiembuisjes wordt algemeen beschouwd als de fenotypische verandering waarbij Candida invasieve eigenschappen gaat vertonen. 

Wilde oregano-olie en vooral carvacrol blijkt in vitro in lage concentraties de activiteit van HLE (Human Neutrophil Elastase) te remmen waardoor oregano-olie mogelijk een fytotherapeuticum kan zijn bij ziekten als COPD en emfyseem.

Carvacrol vertoont in vitro een cholinesterase-remmend effect dat tienmaal sterker is dan van zijn isomeer thymol. De eigenschap om cholinesterase te remmen kan van nut zijn in het tegengaan van cognitieve achteruitgang zoals bij dementie.

Testen en onderzoek naar het gedrag van muizen toont voor carvacrol een anxiolytisch effect bij een eenmalige dosering. Het effect van zowel carvacrol (12,5; 25 en 50 mg/kg) als diazepam (1 en 2 mg/kg) kon met flumazenil (benzodiazepine-antagonist) ongedaan gemaakt worden.

Carvacrol in dezelfde doseringen resulteerde in een antidepressief effect wanneer muizen werden blootgesteld aan stressvolle testen. De werking werd toegeschreven aan een interactie met het dopaminerge systeem en niet aan het serotonerge of noradrenerge systeem.

Biggen die na de zoogtijd voer krijgen dat vermengd is met carvacrol of thymol vertonen in de gehele dunne darm een hoger villus/crypt ratio dan de controlegroep die geen toevoeging kreeg. Dit bleek uit de toepassing van meerdere experimentele voeders. De experimentele voeders bevatten 500 en 2000 mg carvacrol of thymol  per kilogram voer.  Carvacrol en thymol verbeterden in dit onderzoek de gezondheid van de darm hetgeen met histologisch onderzoek werd vastgesteld. In geen enkele darmsectie was echter het aantal bacteriën door carvacrol of thymol verminderd. Als gevolg van de voedselinname met 2000 mg carvacrol per kilogram voer, was de concentratie carvacrol in de maag  en het begin van de dunne darm resp. 521 en 5 mg per kilogram spijsbrij. De resorptie van carvacrol blijkt dus meer dan 90% te zijn.  

Een oregano-olie-emulsie van 600 mg per dag gedurende 6 weken aan 14 patiënten met darmparasieten (Blastocystis hominis, Entamoebe hartmanni en Endolimax nana) resulteerde in een complete eliminatie van Entamoebe hartmanni (4 patiënten), Endolimax nana (1 patiënt) en Blastocystis hominis (8 patiënten). Bij 3 andere patiënten daalde de score voor Blastocystis hominis. Bij 7 van deze 11 patiënten met Blastocystis hominis verbeterden de maag-darmklachten.

In vitro onderzoek toont verder dat carvacrolrijke oregano-olie klinisch van nut kan zijn voor de behandeling van Giardia lamblia. De concentratie van de carvacrolrijke oregano-olie van twee verschillende soorten bedroeg 71 tot 85 microgram/ml en blokkeerde binnen een uur de adhesieve eigenschappen van de parasiet waarbij in de navolgende tijd tevens de helft van het aantal parasieten werd gedood. Een enkel vergelijkend in vitro onderzoek met een bestaand medicijn tegen Giardia (Tinidazole), toont zelfs dat een wilde oregano-olie beter werkt dan Tinidazole (anti-giardia-medicijn). 

INDICATIES

Filosoferend over oregano-olie en de tot dusver waargenomen effecten en mechanismen zou de therapeutische bandbreedte van wilde oregano-olie mogelijk onder meer vertegenwoordigd kunnen worden door een remmend effect op provocaties van bacterieel LPS uit de humane darm via het poortadersysteem op de lever. Dergelijke provocaties via de humane darm zijn deel van het verklaringsmodel voor het persisteren van een laaggradig ontstekingssyndroom als causaal mechanisme voor vele pathologieën als het metabool syndroom, hart- en vaatziekten, diabetes type 2 en diverse aan ouderdom gerelateerde (neuro-)degeneratieve ziekten. Als we daarbij de waarnemingen beschouwen die wijzen op het verbeteren van de gezondheid van de dunne darm en haar villi, alsmede de eigenschap om  Candida en andere darmpathogenen te decimeren, dan kan men niet ontkennen dat er in potentie een grote klinische waarde voor wilde oregano-olie is weggelegd. Daarbij kunnen de specifiek ontstekingsremmende en PPAR-gamma activerende eigenschappen bijdragen aan het verminderen van insulineresistentie en het normaliseren van het glucose- en vetmetabolisme. Meer in vivo studies en klinisch onderzoek naar de toepassing van oregano-olie bij de mens zijn nodig om de medische betekenis verder te kunnen determineren. 

Tot dusver wordt wilde oregano-olie door menig behandelaar geadviseerd bij een gastro-intestinale candidiasis (GIC). Een dergelijke toepassing is gebaseerd op klinische ervaringen. Zo werd in casussen van patiënten met het chronisch vermoeidheidssyndroom een oraal caprylzuurpreparaat beschreven als een initiële behandeling tegen GIC. Vervolgens werd overgegaan op een orale toepassing van een plantaardige olie als wilde oregano-olie om overgroei van Candida te bestrijden in huidplooien, nagels, oren en sinussen. 

In-vitro-onderzoek toont dat  caprylzuur in een hoge dosering het snelst werkt tegen Candida, maar dat laurinezuur superieur is in lage doseringen. De minimale groeiremmende concentratie tegen Candida is echter het laagst voor oregano-olie en alleen voor wilde oregano-olie is in vivo bij muizen een overtuigend systemisch anti-Candida effect aangetoond.

Daarnaast zou wilde oregano-olie van nut kunnen zijn bij de behandeling van een aantal darmparasieten als Blastocystis hominis, Entamoebe hartmanni en Endolimax nana. In-vitro-onderzoek toont ook de antiparasitaire eigenschappen tegen Giardia lamblia. Het onderzoek naar het antiparasitaire effect van wilde oregano-olie is tot dusver nog mager. Kortom, wilde oregano-olie lijkt conform de traditionele toepassing zijn waarde te hebben bij interventies ter sanering van het spijsverteringstelsel en het immuunsysteem. Verder blijkt de olie traditioneel nuttig bij geestelijke uitputting en werkt het kalmerend en versterkend.

BIJWERKINGEN

Oregano en oregano-olie worden beschouwd als een veilig voedingsadditief in Amerika en Europa. Verder zijn er van de oregano-olie geen ongewenste bijwerkingen bekend. 

Oregano-olie met carvacrol als belangrijkste inhoudstof blijkt in acute en chronische preklinische toxiciteitsmodellen niet toxisch te zijn. Carvacrol (5-isopropyl-2-methylphenol) is niet hepatotoxisch voor ratten. Onderzoek naar de afzonderlijke oliefracties van oregano toonde aan dat carvacrol, in tegenstelling tot thymol, niet irriterend is voor de huid. Er zijn enkele gevallen bekend van een systemische allergische reactie als gevolg van inname van oregano. 

De pure etherische olie van wilde oregano kan irriterend zijn voor de slijmvliezen. Derhalve wordt geadviseerd om bij directe toepassing op de slijmvliezen een concentratie van 1% te gebruiken.  

Aangezien de interne dan wel orale therapeutische toepassing van wilde oregano-olie bij mensen nauwelijks onderzocht is, wordt het gebruik ervan tijdens de zwangerschap afgeraden. De Duitse Kommission E hanteert geen advies voor therapeutisch gebruik omdat volgens hen de werking onvoldoende is gedocumenteerd.

Nog over marjolein

Origanum is een geslacht van meer dan 30 soorten uit het mediterrane gebied en het Midden-Oosten. Ze behoren tot de lipbloemenfamilie (Lamiaceae). Alle Origanum-soorten zijn aromatische, droogtebestendige kruiden. 

De wilde marjolein heeft verschillende ondersoorten en dwergvormen en is een winterharde plant. De plant komt in Nederland ook in het wild voor. Het is de “klassieke mediterrane” oregano die bij de meeste mensen in het keukenkastje staat. Het kruid heeft een mild, kruidig aroma, dat in allerlei zomerse gerechten gebruikt wordt.

De echte marjolein wordt ook majoraan genoemd (naar de wetenschappelijke naam). Het is een groenblijvende halfheester met grijsgroen blad en bolvormige hoofdjes van kleine, witte bloemetjes. De soort is zeldzaam in Nederland (zeer sporadisch in de duinen en Zuid-Limburg), maar komt in het wild veel voor in Turkije en Syrië. Echte marjolein lijkt erg op wilde marjolein. Een belangrijk en herkenbaar verschil is dat bij echte marjolein de bladeren geen bladsteel hebben en bij wilde marjolein wel. Echte marjolein heeft een verfijnd aroma, zachter en zoeter dan wilde marjolein. Om de verwarring compleet te maken, wordt soms de naam zoete marjolein gebruikt voor deze soort, omdat de etherische olie uit de plant geen fenolen bevat, in tegenstelling tot wilde marjolein.

Majoraan en oregano zijn dus twee verschillende soorten van dezelfde familie. Daarnaast komen ook andere Origanum-soorten voor, zoals de Syrische marjolein (Origanum syriacum) en de Hopmarjolein (Origanum dictamnus). Alle soorten Origanum passen goed bij citroen, knoflook, marinades, vlees, vis, hartige salades, mediterrane gerechten en sauzen en gerechten op basis van tomaat.

Overigens kun je in andere werelddelen planten vinden die ook “oregano” genoemd worden, maar die botanisch niets te maken hebben met Origanum-soorten. Zo wordt Lippia micromera (familie van de verbena’s of Verbenaceae), die in het wild voorkomt in de zuidelijke Verenigde Staten en Midden-Amerika, ook wel Jamaicaanse of Mexicaanse oregano genoemd (en ongetwijfeld nog tientallen andere namen, naargelang de nationaliteit van de spreker). Vijf-in-een-kruid (Coleus amboinicus) is een andere lipbloemige die oorspronkelijk uit Afrika komt en net als oregano in de keuken wordt gebruikt. Ook deze plant heeft botanisch weinig te maken met Origanum-soorten. De bladeren zijn bijna zo dik als vetplanten en ook de samenstelling van de etherische olie verschilt sterk met die van Origanum.

RECEPT: Marjoleinolie

De wilde en de echte marjolein worden veel toegepast in de mediterrane keuken. Marjoleinolie is een simpel recept om zomerse gerechten op te vrolijken.

Wat heb je nodig voor een flesje van 500 ml?

Een handvol verse blaadjes van de wilde marjolein

Leg de knoflooktenen een paar uur in azijn of citroensap (*). Kneus de kruiden lichtjes, met een mes of in een vijzel. Stop de kruiden in een schone fles en voeg de olijfolie toe. Laat het 2-3 dagen trekken, zodat de olie de smaken kan opnemen. Bewaar het koel en donker voor maximaal twee weken.

(*) Knoflook in olie wordt nogal lekker gevonden door de bacterie Clostridium botulinum, die botulisme kan overbrengen. Botulisme is dodelijk. Het gevaar schuilt in de toxine (het gif) botuline dat de bacteriën kunnen uitscheiden. Dit is één van de gevaarlijkste gifstoffen die we kennen. Zorg dus voor een ongunstig milieu om de groei van Clostridium Botulinium tegen te gaan. De groei van de bacteriën kan voorkomen of geremd worden door zuren (azijn of citroensap helpen), hoge concentraties aan suikers, een hoog zuurstofniveau, een laag vochtniveau en bewaring aan temperaturen onder de 3 graden. Laat de knoflooktenen daarom eerst marineren in een zuur en bewaar de olie kort en in de koelkast!
