Berkenzwam tegen kanker?

In Siberie oogsten de inheemse telers een zwam van de berk, waarvan ze thee maken, en die thee wordt ingezet bij de bestrijding van kanker. In die zwam zit heel veel betuline zuur, een actieve component van de berkenbast, die via de paddestoel relatief makkelijk oplosbaar is als een soort thee. Nu, die oude gewoontje blijkt bij nader inzien wetenschappelijk bijzonder boeiend te zijn. Het berkenzuur uit de berkenbast dat in die paddestoel zit is een enorm sterk anti-kanker middel.

Betuline is een bijzonder molecuul, een triterpeen die apoptose ofwel celdood veroorzaakt in kankercellen. De anti-kanker werkzaamheid van dit zuur wordt in veel studies duidelijk gemaakt, studies met uiteenlopende modellen. Ook semisynthetische moleculen die afgeleid zijn van betulinezuur werken als cytostaticum.

De paddenstoel op de berkenstam die veel genoemd wordt, heet de chaga, en die paddenstoel zit letterlijk stampvol met anti-oxidanten. Het is de sterkst werkzame plantaardige vorm van anti-oxidanten en deze paddenstoel is dan ook super boeiend als nieuw anti-kanker middel.

Vermoedelijk hebben de sjamanen uit Siberië dan zelfs gelijk als ze thee van deze zwam voorschrijven bij kanker.

Uiteraard zijn er nog geen klinische studies met de zwam die zoveel anti-oxidanten bevat, maar deze plantenvorm is dus uiterst boeiend!

Inonotus obliquus (also known as Chaga) is a fungal parasite of birch trees that has been prized in folkloric medicine for centuries due to its curative powers against cancer and infectious diseases. Over the past decade, a number of studies have examined the anti-tumor properties of various Chaga extracts and demonstrated remarkable efficacy of this natural product in the treatment of solid tumors in mice. We sought to determine if ethanol extracts from Chaga affected the oncogenic properties of cultured melanoma cells. Treatment of B16F1 mouse melanoma cells with fresh ethanol extracts of Chaga resulted in strong increases in melanocyte dendricity as evidenced by increases in the number of cells with dendritic projections, the number of projections per cell, and total projection length. Chaga treatment led to cell cycle disruption as evidenced by decreased steady state cyclin mRNA expression and an increase in the number of cells in the G2 phase of the cell cycle. Moreover, Chaga treatment led to melanoma cell toxicity in a dose dependent manner. These data suggest that ethanol extracts of Chaga may have the potential as an anti-melanoma treatment.

De chaga onttrekt betuline aan de berkenbast en zet dat om in betulinezuur. Voor de talloze gezondheidsclaims die aan de zwam worden toegekend, bestaat geen wetenschappelijk bewijs zegt Jan Paul Medema, hoogleraar experimentele oncologie en radiobiologie in het Amsterdamse AMC. Maar één claim, de meest spectaculaire, blijkt deels wél te kloppen: zuiver betulinezuur doodt inderdaad kankercellen. Of chaga dat ook kan, is echter niet bewezen, zegt hij. Samen met de Leidse immunoloog Jan Kessler doet Medema al een paar jaar onderzoek naar het zuur. Het tumor dodende effect ervan werd al in de jaren negentig ontdekt, zegt hij, en sindsdien zijn talrijke internationale studies gedaan. Alleen: verder dan het laboratorium en proefdieren komen de onderzoekers niet. Medema en Kessler deden onderzoek naar de werking van betulinezuur op cellijnen van borst-, prostaat-, darm-, long- en baarmoederhalskanker, en publiceerden daarover in Cancer Letters. Betulinezuur blijkt op meerder manieren de celdood van tumorcellen te bewerkstelligen en laat gezonde cellen ongemoeid, zegt Medema. Groot probleem is echter dat betulinezuur slecht oplost in water, waardoor het lastig is toe te dienen. Omdat het zuur wel oplost in vet, wordt geprobeerd het in vetbolletjes te verpakken zodat het mogelijk makkelijker wordt opgenomen in het lichaam. Bij muizen die werden gevoed met in maïsolie opgelost betulinezuur, werden goede resultaten geboekt, zegt Medema. ‘Maar als we dat bij mensen willen doen, moeten we zulke grote hoeveelheden geven. Dat is niet haalbaar.’ Klinisch, gerandomiseerd onderzoek bij mensen ligt niet in het verschiet, denkt hij. ‘Die studies zijn erg duur en er valt voor een commerciële partij, zoals een farmaceutisch bedrijf, weinig te halen omdat betulinezuur een natuurproduct is en nu al vrij verkrijgbaar is.' Dat kankerpatiënten elkaar het middel aanbevelen, vindt hij begrijpelijk maar de wonderbaarlijke genezingen die eraan worden toegeschreven, zijn onwaarschijnlijk en zeker niet wetenschappelijk bewezen, zegt hij. Medema ziet meer heil in het achterhalen van het werkingsmechanisme van betulinezuur. ‘Waarom pakt het selectief de tumorcellen aan? Als we dat ontdekken, kunnen we het gebruiken voor het ontwikkelen van nieuwe anti-kankermedicijnen.’Ellen de Visser

Medicinal mushrooms have an established history of use in traditional oriental therapy and nutritionally functional foods. Inonotus obliquus (Chaga mushroom) belonging to the family Hymenochaetaceae of Basidiomycetes, preferably grows on the trunks of mature live birch trees [9]. The extracts of I. obliquus have been used in China, Korea, Japan, Russia, and the Baltics for their favorable effects on lipid metabolism and cardiac function, as well as for anti-bacterial, anti-inflammatory, anti-oxidant, and anti-tumor activities [9]. I. obliquus extracts were found to inhibit hepatitis C virus [10] and human immunodeficiency virus [11, 12] and demonstrated strong anti-oxidant and immunostimulatory activities in vitro [13, 14]. At the same time, animal studies revealed that aqueous extracts of I. obliquus exhibited anti-inflammatory effects in experimental colitis [15, 16] and promoted lipid metabolism [17]. Several studies investigated the anti-tumor activity of the I. obliquus aqueous extract and found that it suppressed the proliferation [18] and induced apoptosis [19] of various carcinoma cell lines. Furthermore, the compounds isolated from I. obliquus extracts were shown to inhibit skin carcinogenesis [20] and tumor growth in Sarcoma-180 cell-bearing mice [21]. However, despite increasing evidence of anticancer activity exhibited by the I. obliquus extract and its individual components [9, 22], the underlying mechanisms are still unclear and the effects of I. obliquus on cancer prevention are not understood.
