Het zijn lactonen met een benzeen ring.

Een groep heet: coumarine C9H6O2, met de geur van vers gemaaid hooi.

Coumarine (of cumarine) = 1,2-benzopyron. Het is inderdaad een op zichzelf staande stof, hoewel er diverse andere stoffen afgeleid van cumarine zijn met cumarine in de naam (dihidrocumarine, 6-methylcumarine etc.)

Het is o.m. een geurstof en komt voor in bijvoorbeeld kaneel en tonkabonen. In de parfumerie is cumarine een belangrijke stof, fougère parfums kunnen niet zonder. Coumarine mag niet meer gebruikt worden in levensmiddelen, het heeft een bloedverdunnende werking met alle risico's van dien. Ze komen in de meeste groene groenten voor. Evenals lactonen zijn het sterke bestanddelen: zelfs in kleine percentages hebben ze een sterke therapeutische werking.

Het is een kleurloos, kristalachtig wit, glanzend poeder. Cas-nummer: 91-64-5. Formule: 2H-1-benzopyran-2-one. De geur is van hooi, zoet aromatisch, crème-achtig, met nootachtige tonen, veel gebruikt in de parfumindustrie, zeep en reinigingsmiddelen.

Smeltpunt 71 graden C – kookpunt 301 graden C – vlampunt > 100 graden C. Atoomgewicht: 146,14 g/mol. Oraal giftig: LD50 ratten 293 mg/kg.

Lost slecht op in water, goed in: ethanol, ether, chloroform. De uitgangsstof voor coumarine in planten is kaneelzuur en coumarine is weer de basisstof voor aesculine, umbelliferon en de furocoumarinen.

Ze komen voor in Conyza canadensis 8% - sinaasappel bitter 6% - citroen 4% - bergamot 3% - lavendel 3% - Tagetes 3% - dragon 3% - engelwortel 2% - koriander 2% - dille 2% - grapefruit 2% - lavandin 2% - limoen 2% - zwarte peper 1% - selderij, zaad 1% - kaneel, schors 1% - cistus ladnaiferuas 1% - (cumin) 1% - Artemisia annua (idaho) 1% - melisse 1% - sinaasappel, zoet 1% pepermunt 1% - petit grain 1%.

De naam komt uit het Spaans: camarú en betekent tonkabonenboom.

Coumarinen zijn van phenolzuren afgeleid. Ze bevinden zich veelvuldig in de familie Rutaceae en Apiaceae. Enzymatische wordt coumarine gevormd tijdens het verwelkings- en droogproces en is bijvoorbeeld het aroma van het Onze Lievevrouwe bedstro. Bij het drogen en verwelken komt coumarine vrij uit de suikerketens waar het in de levende plant aan gebonden is.

Dikwijls wordt coumarine in de planten vergezeld van etherische olie.

Vroeger werd het gebruikt als smaakstof, maar werd in 1954 verboden toen de FDA coumarine het predikaat: carcinogeen gaf, met als gevolg dat het niet in voedingsmiddelen gebruikt wordt, terwijl het aanwezig is in veel natuurlijke planten: tonka bonen (1-10% in de bonen, absolute 65%), lavendel (8% in de absolute), lavandin (5% in de absolute),zoethout, abrikozen, aardbeien, cassia 4-11% Burfield 1999), kaneel, klaver, kersen, Melilotus officinalis L. (in gedroogde toestand 0.9%, Wagner 1996 in het kruid 0.25 – 0.45% samen met umbelliferon , scopoletine, enz.), Narcissus species, Artemisia vulgaris L., Michelia champaca L. Artemisia annua L. e. a.

De S.C.C.P. (Scientific Committee on Consumer Products) geeft coumarine aan als allergeen. Cropwatch geeft aan dat hier onvoldoende bewijs voor is en noemt het een zwakke sentisizer. Concluderend naar aanleiding van Cropwatch komt het er op neer dat de SCCP haar eenmaal ingenomen standpunten meer defensief dan objectief bekijkt.

De ontdekking van de stof kwam doordat een aantal koeien gegeten hadden van een partij gebroeide klaver. Deze dieren kregen bloedingen. Bij het drogen van de planten komt de coumarine vrij uit de suikerketens waar het in de levende plant aan gebonden is.

Coumarinederivaten zijn gevaarlijk. Een klein beetje teveel maakt het bloed onstolbaar hetgeen tot levensgevaar kan leiden. Coumarine is een anticoagulant of bloedverdunner en bij inname kunnen leverschade, nierschade en hoofdpijn optreden. Niet teveel kaneel gebruiken. Coumarine heeft een remmende werking op de bloedstolling. Effectief tegengif is vitamine K.

Gebruikt als rattenbestrijdingsmiddel: bij hoge doses treden inwendige bloedingen op. De ratten en muizen overlijden na enige dagen. Omdat het zo lang duurt brengen de andere ratten het vergif eten niet meer in verband met het doodgaan. Bij snelwerkende vergiften wordt dit verband door de ratten/muizen soms wel gezien en stoppen ze met het eten van deze stoffen.

Coumarine wordt synthetisch gemaakt d.m.v. een Perkin synthese (W.H. Perkins 1868) uit salicylaldehyde en ijsazijn. Vroeger gebruikt als smaakstof.

Belangrijk als geurstof in parfum. Wordt ook gebruikt als optische witmaker, vanwege fluorescente eigenschappen van coumarine.

Veel zijn huidirritant en phototoxisch (licht). Voorzichtig gebruiken. In werking zijn ze anti-coagulatief – hypotensief – opwekkend en toch sedatief, ontspannend, krampwerend, antitrombose, photosensitiverend, verlagen de bloeddruk. Sommigen zijn anti-fungal en antiviraal. Anti-convulsief (vermindert stuipen). Bergapteen is potentieel carcinogeen.

Worden gevonden in bergamot, sinaasappel, mandarijn, citroen. In geperste citrusvruchten. Verder in akkerhoningklaver, reukgras, rolklaver, kaneel, tonkabonen. In gedestilleerde olie komen ze zelden voor, omdat hun moleculen te groot zijn. Ze worden gewonnen in de laatste 10 minuten van de destillatietijd, in sporen komen ze dan voor en alleen in complete, hoogwaardige oliën.

Coumarinederivaten werken phototoxisch. De moleculen binden zich aan de melaninecellen in de huid. Deze absorberen daardoor het ultraviolette licht en veroorzaken zo lichte tot sterke verbrandingen, afhankelijk van het huidtype.

De familie van de schermbloemigen en de wijnruit bevatten furocoumarinen, die als fotosensibiliserende stoffen de oorzaak zijn van “lichtziekte” of weide-dermatitis of zwembad-dermatitis. Na contact met het sap van de betreffende planten van berenklauw, engelwortel, pastinaak, duizendblad bijvoorbeeld door het liggen op pas gemaaide weiden, wordt de huid op deze plaatsen overgevoelig voor zonlicht en dan ontstaan rode plekken op de huid, ontstekingen en soms volgt een algehele malaise. Vlekken in de huid die na het gebruik van eau de cologne in het zonlicht optreden, zijn een gevolg van het gehalte aan bergapteen in de bergamotolie. Wegens de giftige bijwerkingen is het niet mogelijk deze algemeen te gebruiken om een bruine huidskleur te krijgen. Wel gebruikt men coumarine, bijvoorbeeld aesculine, uit de paardekastanje in middelen tegen zonnebrand, omdat ze uv-licht van bepaalde golflengten absorberen. Coumarine komt in grote hoeveelheden voor in de tonka-boon en eveneens in akkerhoningklaver, lievevrouwebedstro en in bastaardmelisse.

Alhoewel verboden in de USA, Japan, India en de EU, kan het worden gebruikt als additief in cosmetica, eetwaren en in bepaalde aroma’s in beperkte concentraties. In Duitsland was het verboden van 1970-1991 en dat werd toen veranderd in dee concentratie limiet. Door recent onderzoek nu veel gebruikt in de fytotherapie, bijv oorbeeld in Melilotus-extract.

Puur is het een geel tot beigeachtig cristallijn poeder van de familie coumarine. Reguleert de bloeddruk: van hoog naar lager en vice versa. Heeft bacteriostatisc\he activiteiten tegen Escherichia coli, Staf. Aureus, Streptococcus species, Klebsiella pneumonia en Pseudomonas aeruginosa. Scopoletine heeft anti-inflammatoire eigenschappen en kan worden gebruikt om bronchitis en astma te behandelen. Ook reguleert het srotonine, het hormoon tegen angst en depressies.

Wetenschappelijk Comité voor de voeding van de Europese Unie (FC) geefr aan dat de geisoleerde coumarine uit kaneel in dierproeven kanker kan verwekken en mogelijk ook het erfelijk materiaal aantast (mutageen of gentoxisch). De jongste gegevens wijzen erop dat coumarine bij de mens minder schadelijk is dan bij proefdieren.

Twee geur- en smaakstoffen, die van nature voorkomen in basilicum en nootmuskaat, evenals in venkel, anijs en andere kruiden lijkt op dat van coumarine. Methyleugenol voorkomend in nootmuskaat, dragon, basilicum, steranijs en venkel is carcinogeen en genotoxisch. De stof is verwant aan safrol dat in Europa en de VS al lange tijd verboden is. Ook estragol in dragon, venkel, anijs en basilicum is carcinogeen en genotoxisch. Het wordt als smaakstof toegevoegd aan vetten, oliën, visconserven en alcoholische dranken. De dierproeven met methyleugenol en stragol geven diverse soorten tumoren, vooral leverkanker. Daarom is het advies beperking als smaakstof van deze twee en een matig gebruik van de desbetreffende kruiden. In Duitsland zijn ze reeds verboden als smaakstof en wettelijke regels zijn in de maak betreffende Europa. Het gebruik van etherische olie blijft waarschijnlijk toegestaan.

In de farmacie vormt coumarine de chemische basis van de orale anticoagulantia, of de zogenaamde coumarinederivaten. Coumarine wordt aangewend bij lymfe-oedeem. Coumarine zelf heeft geen anticoagulerende werking. Het vermindert het volume van de oedemen met hoog proteïnegehalte door verhoging van de proteolyse door de marcophagen, waarvan het aantal en de activiteiten verhoogd worden. Bovendien verminderen pijn en ontstekingen, verhoogt de mobiliteit en verloopt het genezingsproces van kwetsuren gemakkelijker.

De hepatotoxiciteit is reeds jaren bekend. Het veroorzaakte effect gaat van de verhoging van de lever enzymgehaltes tot orgaanschade en kan tot de dood leiden.

The variety of physiological effects of natural coumarins is extensive. Humans are exposed to themin everyday life through the handling of garden plants like Aegopodium podagraria or ingestion of fruit, vegetables or spices (e.g. Petroselinum crispum).

Based on the ethnopharmacological background and reported biological activities, properties of some of the natural coumarins have been investigated more thoroughly using either pure compounds or extracts from plants containing them.

The anti-inflammatory screening of twenty coumarin compounds studied with an in vitro model forelastase secretion in human neutrophils using PAF and fMLP as stimuli revealed that the ethnobotanical use of Angelica archangelica, Petroselinum crispum and Ruta graveolens as a relieffor cough, colds and arthritis might be explained by the presence of linear furanocoumarins psoralen and xanthotoxin. Coumarins are considered as phytoalexins since plants produce them as defence substances when wounded or attacked by other organisms.

The antimicrobial effects of methanol extracts prepared from seven plants growing in Finland, namely Aegopodium podagraria, Anethum graveolens, A.archangelica, Levisticum officinalis, P. crispum, and Peucedanum palustre, and R. graveolens, andpure coumarins occurring in them were merely observed against plant pathogens which supported the role of coumarins and furanocoumarins as defensive compounds. The agar-diffusion methods used are suitable for the bioassay-guided isolation of active substances.The phototoxicity of linear furanocoumarins (also referred to as psoralens) has been turned into a useful property when combined with controlled UVA irradiation, this PUVA treatment has been widely used for psoriasis. For screening phototoxic compounds and extracts, also others thancoumarins, a microwell plate test with Artemia salina as test organism was developed. It is noteworthy, that with this test system it is possible to investigate both toxicity and phototoxicity atthe same time with the same concentrations.Furthermore, osthol, a simple coumarin from Angelica archangelica, is suggested to be a usefulcompound for investigations on ligand-receptor interactions and for receptor-mediated regulationsof intracellular free calcium concentrations.

Coumarins can be suggested to be beneficial for the plants themselves as natural biocontrolling antipathogenic compounds, and for humans as remedy for hyperproliferative skin diseases and asreference compounds in various bioactivity tests. Furthermore, coumarin-containing plants arevaluable as dietary supplements on the basis of their mild antimicrobial and anti-inflammatory effects

Natural coumarins, like other unsaturated lactones, may exert various effects on living organisms.The extensive range of physiological effects, both in plants and in animals, in addition to lowtoxicity (with the exception of phototoxicity, which, however, with careful dosimetry is one of themost important indications to this group), relative cheapness, presence in the daily groceries asspices, fruit and vegetables and occurrence in various herbal remedies of coumarins, led us toinvestigate more thoroughly properties of some natural coumarins and coumarin containing extracts with ethnopharmacological background.

Twenty coumarin compounds were studied for their anti-inflammatory properties in an in vitromodel for elastase secretion in human neutrophils using PAF and fMLP as stimuli. The activities ofthe compounds used were relatively modest and non-specific, psoralen, xanthotoxin, ledebouvielloland athamantin being the most active ones. The anti-inflammatory activity may be involved in their ethnobotanical use as remedies for cough, cold and rheumatism (arthritis). Besides anti-inflammatory activity, psoralen and xanthotoxin were active also in the phototoxicity tests, and bothof these activities may support their use in the treatment of psoriasis.

Antimicrobial screening against selected Gram-positive and Gram-negative bacteria, yeasts, mold,as well as plant pathogenic fungi, with emphasis on method optimisation was carried out onmethanol extracts prepared from seven plants growing in Finland. The selected plant species were merely active against plant pathogens which supports the role of coumarins and furanocoumarins asdefensive compounds rather for the plant itself. The agar-diffusion methods (discs, holes) utilisedserve as reference methods when developing a more effective microwell plate test. Microwell technique was already applied in developing an easy-to-perform and inexpensivemethod, the Artemia salina (brine shrimp) larvae test, for screening phototoxic compounds. Linearfuranocoumarins bergapten, psoralen and xanthotoxin were phototoxic and umbelliferone as asimple coumarin was not, as expected along with earlier observations. Methanol extracts from sevencoumarin containing plants were also studied, and the clearest phototoxic activity was observedwith Angelica archangelica throughout the vegetative period and with Peucedanum palustre at laterstages of growth. Additionally, it is possible to investigate also toxicity at the same time with thesame concentrations, as seen with peucedanin and the extract of Ruta graveolens.

Based on earlier observations (VUORELA, 1988; TÖRNQUIST and VUORELA, 1990;HÄRMÄLÄ et al., 1992), columbianadin, osthol and xanthotoxin were used to evaluate further the mode of action on calcium fluxes in rat thyroid FRTL-5 cells as well as in rat pituitary GH4C1 cells. The results suggest that osthol may prove to be a useful compound for investigations on ligand-receptor interaction, and some coumarins may have a dual mechanism of action: in addition to theirearlier documented effects on VOCCs, they may also interact with receptor-mediated signalling events.

In conclusion, coumarin compounds can be suggested to be beneficial for the plants themselves as natural biocontrolling antipathogenic compounds, and for human beings as dietary supplements onthe basis of their mild antimicrobial and anti-inflammatory effects, and as reference compounds invarious bioactivity tests. The use of these compounds as medicinal agents is of importance in thecase of hyperproliferative skin diseases like psoriasis.

CUMARINE

Die von den Phenolsäuren abgeleiteten Cumarine finden sich häufig in Pflanzen der Familien der Dolden- und Rautengewächse.

Die Stammverbindung ist das Cumarin (1-H-1-Benzopyran-2-on). Es wird beim Welkprozess enzymatisch gebildet und macht z.B. das typische Aroma des Waldmeisters aus. Es ist häufig mit den ätherischen Ölen der Pflanze vergesellschaftet.

Toxikologisch bedeutungsvoll sind das durch Fäulnisprozesse z.B. in angeschimmelten Kleeheu entstehende Dicumarol, das zu hämorrhagischen Erkrankungen von Weidevieh führen kann (sweet glover desease) und die als stark leberkanzerogenen Aflatoxine in Schimmelpilzen, die aus Polyketiden gebildet werden.

In der Medizin werden synthetisch hergestellte 4-Hydroxy-Cumarinderivate als orale Antikoagulantien verwendet.

Furano- und Pyranocumarinderivate:

Furanocumarine entstehen aus isoprenyliertem Umbelliferon, wie z.B. Osthenol, unter Bildung eines 5-er Ringes zu Furanocumarinen, unter Bildung eines 6-er Ringes zu Pyranocumarinen.

Cumarin:

Ähnlich wie das Rutosid wirkt Cumarin entzündungshemmend, abschwellend und durchblutungsfördernd.

Oftmals werden bei Venenleiden Zubereitungen aus cumarinhaltigen Pflanzen, wie z.B. der Steinklee, eingesetzt.

Im Tierversuch zeigte Cumarin allerdings auch eine lebertoxische und kanzerogene Eigenschaft. Aus diesem Grunde wird empfohlen, die Langzeitaufnahme von cumarinhaltigen Lebensmitteln wie z.B. zimthaltige Tees, zu begrenzen. Allerdings ist die Übertragbarkeit der Befunde auf den Menschen strittig, da die Toxizität von Cumarin – speziesabhängig – unterschiedlich ist. In einigen Studien konnte sogar ein günstige Effekte bei Tumorleiden nachgewiesen werden.

Bei höheren Dosierungen von Cumarin treten Kopfschmerzen und Benommenheit auf.

Dicumarol:

Dicumarol wirkt antagonistisch zu Vitmain K und hemmt daher die Bildung von Gerinnungsfaktoren in der Leber, wobei dafür die Hydroxylgruppe an C-4 des Cumarin-Grundgerüsts essentiell ist. Aus dieser Verbindung wurden die oralen Antikoagulantien entwickelt.

Furanocumarine:

Besonders lineare Furanocumarine vom Typ des Psoralens besitzen photosensibilisierende Eigenschaften, d.h. es können in Verbindung mit Sonnenlicht starke Entzündungen der Haut und Sonnenbrand hervorgerufen werden.

Dieser Effekt wird medizinisch zur Behandlung von Psoriasis, bei Vitiligo und der PUVA-Therapie (Psoralenderivat + UVA-Bestrahlung) ausgenutzt.

Pyranocumarine:

Visnadin wirkt im Tierversuch erweiternd auf die Herzkranzgefäße und positiv inotrop. Die Wirksamkeit ist jedoch nicht durch klinische Studien belegt.

Aflatoxine:

Aflatoxine werden von bestimmten Schimmelpilzen auf stärke- und fetthaltigen Lebensmitteln, wie z.B. Nüsse, Brot, Mais, gebildet und wirken karzinogen auf die Leber. Verschimmelte Lebensmittel sind daher nicht mehr für den Verzehr geeignet und sollten komplett weggeworfen werden.
