Inhoudsstoffen van geneeskrachtige planten: algemeen kort overzicht

Bitterstoffen. De term “bitterstoffen” staat voor een omvangrijke groep van chemisch zeer verschillende, stikstofvrije, zuurstofhoudende plantbestanddelen die verantwoordelijk zijn voor de bittere smaak van veel planten.

Enkelvoudige bitterstoffen (amara pura) worden in belangrijke mate als maagmiddel gebruikt, om de afscheiding van spijsverteringssappen in de mond, de maag en de darm te bevorderen. Deze stoffen worden bijvoorbeeld aangetroffen in de verschillende maagbitters. Typische voorbeelden uit de gentiaanfamilie (Gentianaceae) zijn gele gentiaan en het duizendguldenkruid (Centaurium spp.)

Scherpe bitterstoffen (amara aromatica) stimuleren de warmte- en/of pijnreceptoren. Daartoe behoren bijvoorbeeld de stoffen in gember (Zingiber officinale) of spaanse peper (Capsicum sp..). Ze verhogen de afgifte van spijsverteringssappen (speeksel, maagsap) en worden daarom gebruikt bij winderigheid en maagklachten. Maar ook de schermbloemigen zoals venkel, anijs en karwij horen bij deze groep)

  • - Oplosbaar in water (thee).

  • - Oplosbaar in alcohol 30%-40%

  • - Werking wordt mogelijk gedeeltelijk te niet gedaan door suiker of honing (smaak wordt verstopt?)

Looistoffen of Tanninen: Looistoffen zijn gecompliceerde, stikstofvrije verbindingen (polyfenolen). Hun belangrijkste eigenschap is de adstringerende (=samentrekkende) werking op de huidweefsels. De toepassing van looistoffen vindt dan ook plaats bij ontstekingsprocessen die zich door roodheid, opzwellen van weefsel en verhoogde afscheiding openbaren. Door samentrekking van de oppervlakte volgt een verdichting van het weefsel; de roodheid neemt af en door het binnendringen van de looistoffen in de slijmklieren vertraagt de secretie. Bovendien hebben looistoffen in zekere mate antiseptische, desinfecterende en enigszins lokale anaesthetische eigenschappen. Uitwendig vinden looistofbevattende kruidenbereidingen toepassing als mond- en gorgelwater. Inwendig dienen looistofhoudende kruiden voor de behandeling van diarree. Vroeger werden looistoffen gebruikt bij het looien van dierenhuiden. Planten die looistoffen bevatten zijn de theeplant (Camellia sinensis), de eik (Quercus spp.) en blauwe bosbessen (Vaccinium myrtillus).

Tannines zijn polyfenolen. De stof is van invloed op de smaak, kleur en structuur van onder andere wijn en thee. De naam stamt van het Latijnse tannare, dat looien betekent, een procedé waarmee van huiden leer wordt gemaakt. Ze zijn opgelost in het celvocht van de plant of geconcentreerd in speciale holten met celvocht (looistofvacuolen). In zieke of door parasieten aangetaste planten kan men hoge concentraties van deze stoffen aantreffen.

    • Doordat ze een verbinding met eiwitten aangaan, leggen ze een licht membraan op het slijmvlies. Dit vlies trekt samen waardoor de zwelling, roodheid en overmatige secretie wordt verminderd en uitgezette capilairen worden samengetrokken  bloedstelpend.

    • Het membraan houdt bacteriën tegen  antiseptisch

    • Verstevigende werking op weefsels.

    • Bij roodheid, zwelling (ook klierzwelling), blaasjes, blaren, ontstekingen en bloedingen.

De tannines kunnen in twee groepen onderverdeeld worden:

  • - Hydrolyseerbare tannines, zoals looizuur (verbindingen die splitsen onder invloed van water)

  • - Niet-hydrolyseerbare tannines

In de voeding gaat men

  • - Zout of iets bitters aan het gerecht toevoegen om de tannine van de wijn weg te werken.

  • - Suiker in een gerecht gaat de tannines van de wijn juist sterker naar voor brengen.

Oplosbaarheid en interacties

  • - Doen oplosbare eiwitten neerslaan opgeloste looistoffen worden gebonden en geïnactiveerd door melk

  • - Werking wordt door alkaloïden en zuurstof teniet gedaan

  • - Oplosbaar in glycerine en water

  • - Oplosbaar in alcohol + 50%

Glycosiden. Deze stoffen, die men ook wel glucosiden noemt, zijn glucoseverbindingen die met zuren, basen en enzymen het splitsingsproduct suiker geven. De overblijvende verbinding (aglycon) bepaalt het al of niet werkzaam zijn van het glycoside. Er bestaan verschillende soorten glycosiden, waarvan sommigen sterk werken en zeker niet onschuldig zijn (denk bijvoorbeeld aan digitoxine). Een aantal voorbeelden met specifieke eigenschappen:

Isothiocyanaatglycosiden (mosterdolieglycosiden), die stimulerend werken op de spijsvertering bij anorexie en dyspepsie. Planten met mosterdolieglycosiden zijn bijvoorbeeld herderstasje, witte mosterd, echt lepelblad, mierik en witte waterkers.

Blauwzuurglycosiden, zoals het giftige amygdaline in bittere amandelen.

Anthraquinonglycosiden (emodineglycosiden) zijn afgeleid van 1,8-dihydroxyanthraquinone (dantron, chrysazine), de oxygroepen bevinden zich niet aan één kern. Ze komen voor in rhamnus-, rheum-, cassia-, en aloësoorten. Sporen komen tevens voor in andere planten van de duizendknoopfamilie. Het zijn dikkedarmrelaxantia, die via de dunne darm worden geresorbeerd en in de dikke darm worden uitgescheiden. Aldaar werken ze sterk laxerend door enerzijds verhindering van de waterresorptie en anderzijds reflectorische prikkeling van de dikke darm peristaltiek.

Rubiaglycosiden zijn afgeleid van 1,2-dihydroxyanthraquinone of 1,3-dihydroxyanthraquinone (bijvoorbeeld alizarine), de oxygroepen bevinden zich uitsluitend aan één kern. Zoals de naam al aangeeft komen ze voor in Rubia tinctorum (Meekrap), maar ook enkele andere planten uit de Rubiaceae (sterbladigenfamilie) bevatten deze stoffen, zij het in mindere mate. Voorbeelden zijn geel walstro, kleefkruid en lievevrouwebedstro.

Aucubineglycosiden: komt voor in enkele planten uit de helmkruidfamilie (Scrophulariaceae) evenals in de weegbreefamilie (Plantaginaceae). De farmacologische werking vertoont in verschillende opzichten overeenkomsten met die van de saponinen. Voorbeelden van planten met aucubineglycosiden: ogentroost (Euphrasia officinalis), mannetjesereprijs (Veronica officinalis) en smalle weegbree (Plantago lanceolata).

Fenolglycosiden of arbutosiden: Hiertoe behoren arbutine en de salicylglycosiden. Arbutine is een bestanddeel van de bladeren van de berendruif (Arctostaphylos uva-ursi) en bosbessenplant en wordt meestal via de nieren volkomen onveranderd uitgescheiden. Alleen in alkalische urine wordt arbutine gesplitst tot hydroquinonen, dat een antiseptische werking in de urinewegen heeft.

Salicylglycosiden: Salicine (dat in het lichaam wordt omgezet in salicylzuur), populine en gaulterine hebben praktisch dezelfde farmacologische eigenschappen, namelijk

    • Antipyretisch, analgetisch en antireumatisch

    • Desinfecterend op de slijmvliezen van het maagdarmkanaal

    • Het gezamenlijk glycosidencomplex van de zwarte populier (Populus nigra) namelijk (salicine en populine) verhoogt de oplossing en uitscheiding van urinezuren.

    • Diuretische werking van gaulterine in moerasspirea (Filipendula ulmaria)

Salicylzuren komen voor in onder meer de schietwilg (Salix alba), moerasspirea (Filipendula ulmaria), zwarte populier (Populus nigra)....

Flavonoïd-glycosiden: Deze grote groep bevat vele glycosiden. Het zijn glycosiden die bij hydrolyse als aglycon een flavon geven. De therapeutische werking van de flavonglycosiden is:

    • Algemeen cardiotonisch, vermindering van de permeabiliteit van de capillairen

    • Bevordering van de diurese

    • Vermindering van de galsecretie en mild laxerend

    • Bevordering van de kalkafzetting vanuit het bloed naar de weefsels

    • Algemeen roborerend en toniserend (versterkend, vitamine-werking)

Planten met flavonglycosiden als hoofdbestanddeel zijn meidoorn (Crataegus oxycantha), es (Fraxinus excelsior), kattendoorn (Ononis spinosa), sleedoorn (Prunus spinosa), wijnruit (Ruta graveolens), herderstasje (Capsella Bursa Pastoris), Sint Janskruid (Hypericum perforatum), boekweitplant (Fagopyrum)....

Oplosbaarheid in het algemeen:

  • - oplosbaar in alcohol 30 – 40 %

  • - oplosbaar in water

Anthocyaanglycosiden: Deze kleurstofglycosiden zijn nauw verwant met de flavonglycosiden. Hun fysiologische werking is gelijk aan die van de flavonglycosiden, maar soms zwakker. Planten met anthocyaanglycosiden zijn korenbloem (Centaurea cyanus), groot kaasjeskruid (Malva sylvestris), boerenpioen Paeonia officinalis), grote klaproos (Papaver rhoeas), hondsroos (Rosa canina), maarts viooltje (Viola odorata), Driekleurig viooltje (Viola tricolor).

Saponinen of saponisiden bezitten de eigenschap om in combinatie met water sterk te gaan schuimen. Op basis van hun effect op de oppervlaktespanning hebben ze het karakter van zeep. Rechtstreeks in de bloedcirculatie gebracht werken saponinen als sterk vergif en leiden ze tot oplossing van de rode bloedcellen en tot beschadiging van organen. Grote hoeveelheden veroorzaken krampen en na korte tijd ademverlamming. In het maag-darmkanaal worden saponinen bijna niet opgenomen en ze werken daarom bij opname door de mond in lage doses niet giftig. Saponinehoudende planten worden in de fytotherapie als urinedrijvend middel (berkenbladeren) toegediend, alsook om hun afscheidende en slijmoplossende werking bij bronchitis (sleutelbloem). De wortel van de sleutelbloem (Primula spp.), klimopblad (Hedera helix), (berkenblad (Betula spp.)), zoethout (Glycyrrhiza glabra) en witte paardenkastanje (Aesculus spp.) bevatten saponinen.

Slijmstoffen: Tot de slijmstoffen, die de slijmcellen van de planten bevatten, behoren zetmeel, gom en pectine. Deze worden in de eerste plaats ingezet bij ontstekingen van de slijmvliezen (maag, darm, luchtwegen). De slijmstoffen vormen op de ontstoken slijmvliezen een beschermend laagje. Tot de planten die rijk zijn aan droge slijmstoffen behoren heemstwortel (Althea officinalis), vlozaad (Plantago psyllium), lijnzaad (Linum usitatissimum) en smalle weegbree (Plantago lanceolata).

    • Vooral oplosbaar in water!! Aangezien slijmstoffen vaak gevoelig zijn voor warmte, worden aftreksels van planten in koud of lauw water

    • gemaakt

    • Alcohol kan gebruikt worden als conserveermiddel (max 20%)

    • Zwellen op in koud water en vormen een gelei!

    • Tincturen met laag alcoholgehalte 20° alcohol, wat hoger kan soms wel.

Alkaloïden: zijn stikstofhoudende, organische verbindingen met een complexe structuur. Vaak zijn het sterk werkende stoffen die voorkomen in planten die wij aanduiden met “giftig”, omdat zij inwerken op het centrale zenuwstelsel en kalmerend, pijnstillend, maar ook prikkelend, stimulerend of opwekkend kunnen werken. Tot de alkaloïden behoren bijvoorbeeld morfine en codeïne, die zich in het melksap van de bolpapaver (Papaver somniferum) bevinden, of atropine uit wolfskers (Atropa belladonna), cafeïne in de Koffieplant (Coffea spp.) en nicotine in tabaksbladeren (Nicotiana tabacum). In hoge doseringen zijn diverse alkaloïden toxisch.

    • goed oplosbaar in alcohol 50 – 70 % en ether

    • beperkt oplosbaar in water

    • Azijn kan de werking verhogen, vandaar dat er vroeger wijn en azijn werd gebruikt voor medicinale wijnen en in de wondverzorging

    • onstabiel bij verhitting

    • toxiciteit geneutraliseerd en de werking verminderd een beetje door looistoffen (tannines)

Harsen: Harsen zijn niet-vluchtige stoffen die worden gevormd in de schors en het hout van met name tropische boomsoorten. Wanneer harsen oplossen in vluchtige (etherische) olie, dan wordt het balsem genoemd. Harsen bevatten en zuur en werken daarom prikkelend op de huid. Tot de harshoudende droge stoffen behoren mirre en wierook. Beide werken desinfecterend en ontstekingsremmend en worden vaak toegepast bij ontstekingen van de huid, de keel, de darm of de gewrichten.

    • Oplosbaar in alcohol (70 % – 95%) en warme olie

    • weinig of niet oplosbaar in water

Etherische olie:

Etherische olie wordt ook wel aromatische, vluchtige of essentiële olie genoemd. De etherische oliën zijn vluchtige oliën gewonnen uit planten. Etherische oliën kunnen uit verschillende delen van een plant worden gewonnen, bijvoorbeeld de bloesem, de vrucht, het zaad, de bladeren, de schil, of het hout(van stam, takken of wortels). Soms bevatten verschillende delen van een plant verschillende etherische oliën.

  • - Oplosbaar in alcohol (70 % – 95%) en warme olie

  • - Niet oplosbaar in water

  • - De meeste etherische olie wordt gewonnen door middel van stoomdestillatie (via een alambic). Hierbij wordt stoom door de te verwerken plantendelen geleid. De etherische olie vervliegt met de stoom. Nadat de stoom is afgekoeld tot water kan de etherische olie hiervan worden gescheiden door het verschil in polariteit tussen de olie en het water.

  • - Een methode die bij citrusvruchten gebruikt wordt is de koude persing. Hierbij wordt de oliehoudende schil van de plant uitgeperst en wordt zo de olie gewonnen.

  • - Uit enkele, voornamelijk harsachtige stoffen, wordt de etherische olie gewonnen door droge destillatie. Hierbij wordt de vaste stof voorzichtig verwarmd, waarbij de etherische olie verdampt. Deze damp wordt vervolgens gecondenseerd.

Andere methodes zijn:

  • - extractie, maar strikt genomen ontstaat dan geen etherische olie, maar een extract, absolue of concrète

  • - enfleurage, hierbij ontstaat eigenlijk een pommade (vet) in plaats van een etherische olie. Deze methode wordt niet of nauwelijks nog toegepast vanwege de hoge kosten

Etherische olie wordt op verschillende manieren gebruikt:

  • - Als smaakstof. Vele levensmiddelaroma's zijn geheel of gedeeltelijk opgebouwd uit etherische olie. Voorbeelden zijn sinaasappelolie voor sinas en andere limonades. Pepermuntolie voorde snoepgoed en kruidnagelolie in soepen en sauzen.

  • - Als geurstof. In cosmetica, schoonmaakmiddelen, wasmiddel, Massageolie en luchtverfrissers worden veel etherische oliën gebruikt. Een geurstof voor gebruik in cosmetica bestaat gemiddeld voor 30% uit etherische olie. Daarnaast wordt etherische olie onvermengd of vermengd gebruikt als te verdampen geurstof in een aromabrander, in de sauna of een stoombad.

  • - Als geneesmiddel. Er zijn een paar honderd soorten etherische oliën die gebruikt worden voor hun al dan niet vermeende geneeskrachtige werking. Met name in de aromatherapie -die helemaal gebaseerd is op het gebruik van etherische olie- maar ook daarbuiten.

  • - Als conserveermiddel. Sommige etherische oliën hebben een conserverende werking, bijvoorbeeld wintergreenolie.

  • - Als industriële grondstof. Sommige etherische oliën bevatten een groot gehalte aan een bepaalde stof, zodat het loont om deze daaruit te winnen. Zo wordt eugenol, een grondstof voor vanilline, uit kruidnagelolie gewonnen. Limoneen, dat als verfafbijtmiddel wordt gebruikt, wordt uit sinaasappelolie gewonnen.

Werking:

Zeer verschillend afhankelijk van de plant:

    • Antiseptisch (ontsmettend)

    • Antibiotisch / antibacterieel

    • Ontstekingsremmend

    • Wondhelend

    • Spijsverteringsbevorderend

    • Slijmoplossend

    • Bloeddruk harmoniserend

    • Kalmerend / opwekkend

Mineralen: Verschillende mineralen komen in zeer kleine, maar daarom niet minder belangrijke, hoeveelheden in planten voor. Te denken valt aan natrium, kalium, calcium, magnesium, zwavel, silicium, ijzer, mangaan, koper, zink, fosfor, broom en jodium.

A

Acetylcholine

(vaak afgekort tot ACh) is een neurotransmitter, die vooral betrokken is bij de prikkeloverdracht van de zenuw naar de skeletspiercellen. Het actiepotentiaal maakt in het uiteinde van een zenuwcel acetylcholine vrij. Dit acetylcholine diffundeert door de ruimte tussen deze cel en de volgende cel (deze ruimte heet de synaps) en activeert de laatste door zich te binden aan de acetylcholine-receptor. Na activatie hiervan wordt, als dit een tweede zenuwcel is,weer een elektrisch signaal opgewekt; als het een spiercel is, wordt door de receptoractivatie de spiercontractie (samentrekking) geïnduceerd. Na korte tijd wordt het acetylcholine in de synaptische ruimte door het enzym acetylcholine-esterase razendsnel afgebroken tot de metabolieten choline en acetaat dat in de originele zenuwcel weer tot acetylcholine hergebruikt wordt.

Diverse stoffen en farmaca kunnen dit proces op verschillende manieren beïnvloeden:

1. Agonisten: dit zijn stoffen die de werking van ACh nabootsen, de afgifte van ACh stimuleren of de afbraak van ACh remmen. Bekende ACh-agonisten zijn de zenuwgassen TABUN, SARIN en SOMAN. Hoewel deze stoffen in beginsel de werking van ACh versterken doordat ze de afbraak remmen, werken ze toch op termijn spierverslappend omdat ze een zogenaamd depolariserende verslapping induceren. Daarnaast hebben ze effecten op meerdere andere orgaansystemen.

2. Antagonisten: dit zijn stoffen die de werking van ACh remmen of tegengaan. Belangrijke stoffen zijn atropine (in Wolfskers, Doornappel) en spierverslappers zoals curare (pijlgif van Indianen) en analogen.

De ACh-concentratie is hoog in het corpus striatum en de thalamus maar laag in het cerebellum.

Er zijn drie typen ACh-receptoren:

1. Muscarinische (AChM). Deze zitten veel in de parasympatische eindsynapsen.

2. Nicotinische (AChN). Deze zitten veel in de sympathische en parasympathische ganglia

3. Nicotinische, met receptoren op de motorische eindplaten van skeletspieren (AChNm)

Aescine, triterpeensaponine

Aescine verbetert de bloedcirculatie in de aderen. Aescine zorgt voor een normale wandspanning in de aderen, waardoor het bloed beter naar het hart terugstroomt. Aescine is ook ontstekingsremmend en vermindert oedeem (zwellingen met veel vocht) na trauma's, zeker na sportblessures en operaties.Aescine, het belangrijkste bestanddeel van Aesculus hyppocastanum is een triterpeenglycoside met anti-oedemateuze, anti-exsudatieve, anti-inflammatoire en venotoniserende eigenschappen.In dierexperimenteel onderzoek is gebleken dat het aescine een anti-exsudatief en een vaatwandversterkend effect heeft. Het aescine remt de activiteit van bepaalde lysosomale enzymen, waarvan de activiteit bij chronisch veneus vaatlijden juist is verhoogd, waardoor de afbraak van mucopolysacchariden in de capillairwanden wordt geremd. De filtratie van laag-moleculaire eiwitten, electrolyten en water naar de interstitiële ruimte wordt geremd door een afname van de vasculaire permeabiliteit. In humaan-farmacologisch onderzoek is in vergelijking met een placebo een significante reductie van de transcapillaire filtratie vastgesteld. Tevens is een significante verbetering waargenomen van de symptomen van chronische veneuze insufficiëntie (moeheid, gevoel van zwaarte en spanning in de benen, jeuk, pijn en zwelling van de onderbenen) in verschillende gerandomiseerd dubbelblinde of cross-over studies.

Aescine 2: Antisecretory and gastroprotective effects of aescine in rats.Author: Marhuenda, E : Alarcon de la Lastra, C : Martin, M JCitation: Gen-Pharmacol. 1994 Oct; 25(6): 1213-9Abstract: 1. This study was designed to determine the antisecretory effects of aescine in the perfused stomach of the anaesthetized rat. In addition, the effects of aescine on mucosal lesions produced by intragastric instillation of 1 ml of absolute ethanol, its action on the production of mucus and the possible role of PGs in aescine induced gastroprotection were also studied. 2. Pretreatment of aescine (10 and 50 mg/kg) inhibited the increases in acid secretion induced by histamine (5 mg/kg) and carbachol (10 micrograms/kg). At the highest dose used abolished nearly the increase induced by carbachol (P less than 0.001). 3. Aescine (10, 25 and 50 mg/kg) was found to be effective in the prevention of gastric ulceration induced by absolute ethanol in rats. The degree of gastroprotection decreased with time, the optime effects occurring 60-120 min after oral administration. Pretreatment with indomethacin (10 mg/kg) partially inhibited the gastric protection but the PGE2 determination did not show an increase in prostanoid levels. Furthermore, the protective effect was not associated with an increase in the amount of gastric mucus and glycoprotein content. 4. These results indicate that aescine exerts an antisecretory action which could play a possible role in its antiulcerogentic activity. Also it shows a marked protective mucosal activity which could be partly explained through non-prostaglandin dependent mechanisms involving its antiinflammatory and vasoactive properties.

Alcaloïden / Alkaloïden

Een alkaloïde is een stikstof bevattende plantenbase, met in het algemeen een ingewikkelde chemische (heterocyclische) structuurformule die vaak een sterke fysiologische of farmacologische werking heeft. Sommigen worden als genotmiddel gebruikt. Enkele voorbeelden van alkaloïden die in de geneeskunde worden toegepast zijn: atropine, cafeïne, morfine en codeïne, cocaïne en neostigmine.

Een aantal farmacochemici hanteren een nauwere definitie voor alkaloïden. Naast de hierboven genoemde criteria, stellen zij ook de eis dat de stoffen in zuivere vorm vast moeten zijn, en dat het stikstofatoom deel van de heterocyclische ring moet uitmaken. Op deze wijze gedefinieerd behoren een aantal farmaca van plantaardige oorsprong die ook een sterke farmacologische werking hebben, dan niet tot de alkaloïden. Enkele voorbeelden hiervan zijn colchicine (heeft geen stikstof bevattende heterocyclische ring), nicotine (is een vloeistof) en efedrine (is niet heterocyclisch).

De geschiedenis van de alkaloïden begint in 1806 met de isolatie van morfine uit opium door de apotheker Friedrich Sertürner uit Paderborn. De grote doorbraak hierbij was dat men aanvankelijk naar zure plantaardige stoffen zocht, maar uiteindelijk bleek dat alkaloïden basen waren. De naam alkaloïde werd in 1819 door de apotheker Wilhelm Meissner geïntroduceerd. De naam verwijst naar het feit dat de verbindingen oplosbaar zijn in waterige oplossingen van zuren - ze kunnen zuur neutraliseren - maar niet in waterige alkalische oplossingen.

Vaak hebben planten behorend tot dezelfde familie of hetzelfde geslacht een aantal chemisch verwante alkaloïden. De nachtschadefamilie, verwanten van de aardappel, kent veel soorten die alkaloïden bevatten, zoals de doornappel, het bitterzoet, de zwarte nachtschade, de wolfskers en de groene delen van de aardappel zelf.

Men kan de alkaloïden in een aantal families indelen, waarvan een aantal weer kunnen worden onderverdeeld:

* Fenylalanine familie

1. benzylisoquinoline alkaloïden: papaverine, curare (pijlgif)

2. aporfine alkaloïden: apomorfine

3. berberine alkaloïden: berberine (Berberis vulgaris, Chelidonium)

4. ftalide-isoquinoline derivaten: noscapine

5. morfinaan alkaloïden: codeïne, morfine (Papaver..)

6. restgroep: capsaïcine (Rode peper), emetine

* Tryptofaan familie

Alfa-linoleenzuur

Alfa-linoleenzuur is een essentieel vetzuur voor mensen en zoogdieren, wat betekent dat het lichaam het niet zelf kan maken. Voor optimale groei en conditie is 0,5 procent van de energie-inname per dag nodig, wat voor een volwassene neerkomt op ongeveer 1,5 gram per dag. Er is nog één ander essentieel vetzuur, linolzuur.

Linoleenzuur (CH3CH2CH=CHCH2CH=CHCH2CH=CH(CH2)7COOH) heeft 18 koolstofatomen en drie onverzadigde bindingen tussen koolstofatomen 3-4, 6-7 en 9-10, geteld vanaf het koolstofatoom op het einde (CH3-). Vandaar de benaming n-3 vetzuur of omega 3 (ω3) vetzuur. Notatie is C18:3 n-3. Er is ook nog het gamma-linoleenzuur dat uit linolzuur wordt gemaakt. Dat is een n-6 vetzuur.

Uit linoleenzuur worden de lange-keten n-3 vetzuren met 20 en 22 koolstofatomen gemaakt (onder andere cervonzuur), die belangrijke bouwstenen zijn van membranen in onder andere de hersenen en retina. Linoleenzuurdeficiëntie is nadelig voor de hersenen, ogen en het gehoor. Linoleenzuurdeficiëntie komt echter nauwelijks voor, omdat de voeding voldoende linoleenzuur en langketen n-3 vetzuren bevat. Wat het lichaam aan linoleenzuur niet nodig heeft wordt verbrand en niet opgeslagen in vetweefsel. Dit in tegenstelling tot linolzuur dat wel wordt opgeslagen in vetweefsel. Verder kan het lichaam de lange-ketenvetzuren die uit linoleenzuur worden gemaakt, gedeeltelijk vervangen door lange-ketenvetzuren die uit linolzuur worden gemaakt zonder dat dit tot functionele stoornissen leidt. De gezondheidsclaims van extra linoleenzuur in de voeding zijn vaag en niet goed wetenschappelijk bewezen.

Linoleenzuur zit in sommige plantaardige oliën waaronder rijstolie, walnoten en in geringe mate in groene bladgroenten. Er is ook een type saffloer met zaden waarin linoleenzuur voorkomt. De lange-keten n-3 vetzuren, die het lichaam uit linoleenzuur maakt, zitten in vlees, vis en visolie.

Overview:

Allantoin is a natural compound concentrated in comfrey root that promotes wound healing, speeds up cell regeneration and has a skin-softening (keratolytic) effect. The Merck Index lists the therapeutic applications of allantoin as a topical vulnerary (wound healer) and treatment for skin ulcers. The FDA has approved allantoin skin creams (0.5% to 2.0%) as non-prescription drug products for: 1) the temporary protection of minor cuts, scrapes, burns and sunburn; 2) preventing and protecting skin and lips against chapping, chafing, cracking and wind-burn, and 3) relieving dryness and softening cold sores and fever blisters. Allantoin is also recommended for treating and preventing diaper rash and additionally helps to seal out wetness. Allantoin is also used in shampoos, foam baths, baby powders, lipsticks, various dental preparations and topical pharmaceuticals. The FDA OTC (over-the-counter Drug) Panel does not recognize allantoin as a wound-healing agent, only as a skin protectant. However, allantoin is ter ed as a cell proliferant and epithelization (skin growth) stimulant in texts including the "United States Dispensatory", "Merck Index", and "British Pharmaceutical Codex". Allantoin is said to clean away dead (necrotic) tissue and hasten the growth of new healthy tissue. Since allantoin stimulates new and healthy tissue growth, skin formation may take place over wounds and sores. Allantoin has also been termed a counter irritant that helps alleviate the skin-irritation effects of certain cosmetic ingredients including soaps, detergents, surfactants, oils, and acidic or alkaline materials. Allantoin produces its desirable effects by promoting and speeding up the healthy, natural processes of the body. It is said to help the skin to help itself. Allantoin is the soothing and healing constituent in comfrey root, valued for use in creams and lotions for these properties. Comfrey teas containing allantoin are also recommended for speeding the healing of bruises, sprains, bone fractures and broken bones.

Chemistry:

Allantoin, in its pure form, is a white, odourless, crystalline powder, soluble in water and alcohol and almost insoluble in ether. The Merck Index describes allantoin as a product of purine metabolism that is industrially prepared synthetically by a process using uric acid. It is nontoxic, nonirritating and non-allergenic. Chemical name: glyoxyl-diureide. Molecular weight: 158.12. Melting range: 225C Heavy metals: 10 ppm maximum. Solubility in water: 0.5% at 25C.

Suggested Amount:

Use rate of allantoin is 0.5 - 2.0% w/w. Small concentrations can be added to the water phase, to incorporate more than 0.5% into an emulsion, add during the cooling phase once the temperature has dropped below 50°C/122°F.

Scar development was investigated in 45 young patients who had undergone thoracic surgery. Patients were randomly assigned either to a group which was treated topically with Contractubex gel (Merz + Co., D-Frankfurt/Main), containing 10% onion extract, 50/U of sodium heparin per one g of gel and 1% allantoin, or to a group receiving no treatment. The treatment began on average 26 days after the operation and was continued for one year. The scars of all treated and untreated patients were evaluated at monthly intervals. The appearance of the scar, including scar type and scar size as well as scar colour, was assessed by the physician. A reduction of the increase of scar width was seen in the Contractubex-treated group as compared with the untreated group. Further, physiological scars and skin-coloured scars were more frequent in the treated group than in the untreated group. Hypertrophic or keloidal scars were less frequent in the treated group. No differences in scar length and scar height were seen. At the end of the observation period, the clinical course of scar development was rated as "very good" or "good" in more than 90% of the treated patients, "good" in less than 40% and "moderate" or "bad" in more than 60% of the untreated cases. The tolerability of the drug was "good" or "very good" in all cases. In conclusion, Contractubex gel is useful in scar treatment after thoracic surgery.

Samples which contain 2% (w/w) of allantoin in various emulsified vehicles were prepared and characterized. The influence of vehicle on releasing and diffusion of allantoin through semipermeable membrane into an aqueous medium was examined and the quantity of released allantoin was estimated spectrophotometrically. The best results were achieved with ambiphilic vehicle emulsified with complex Tagat S-Tegin M which in the aqueous phase contained propylene glycol (sample A2). On the contrary, with both lanacolic vehicles, the poorest results were achieved in vitro. On the basis of the results for clinical evaluation, the best preparation was chosen (A2) and the preparation with lanacolic vehicle which contains propylene glycol (B2). Before the application of samples, each patient was tested for irritation and sensitization. All test were negative. During further clinical examinations on patients suffering from psoriasis an open double trial for the duration of 14 d was performed. For the evaluation of the efficacy of the examined preparations, objective parameters of the clinical picture were observed (the state of hyperkeratosis, of erythema and infiltration) as well as subjective parameters which were evaluated by the patients themselves. When the in vitro results are compared with clinical estimation, it is evident that they correspond only when characteristics of the preparation are estimated by patients (smearing, absorption and feeling on the skin), because the best preparation was in these cases A2. Both preparations are equally good when regression of subjective symptoms is evaluated (itching and burning). In the objective evaluation of the regression of visible symptoms, such as hyperkeratosis and erythema, results of the clinical experiment do not correspond with results in vitro.

Allicine, thiosulfinaat in Allium sativum

Synonyms: Diallyl thiosulfinate

Descrition: Allicin is garlic's defence mechanism against attacks by pests. When the garlic plant is attacked or injured it produces allicin by an enzymatic reaction. The enzyme alliinase, converts the chemical alliin to allicin, which is toxic to insects and microorganisms. The antimicrobial acivity of allicin was discovered in 1944 by Cavallito. Purified allicin is not sold commercially because it is not stable and has an offensive odour. Allicin extracted from garlic loses its beneficial properties within hours and turns into other sulphur containing compounds. Diallyl trisulfade, which is similar to allicin but is chemically produced, is stable and is used for treatment bacterial, fungal and parasitic infections.

Distribution: Allicin is the predominant thiosulfinate in garlic (Allium sativum). Allicin is the chemical responsible for he typical and offensive odor of garlic.

Action of Allicin: The antimicrobial effect of allicin is due to its chemical reaction with thiol groups of various enzymes.

The incidence of gastric ulcers is lower in populations with high garlic intake. Studies have confirmed that allicin has inhibitory activity on Helicobacter pylori, a bacteria responsible for the development of gastric ulcers. The sensitivity of Helicobacter pylori to allicin might also explain the lower risk of stomach cancer in people with high garlic intake. A study by Ankri et al (1997) demonstrated that allicin inhibited the ability of the parasite Entamoeba histolytica to destroy monolayers of baby hamster kidney cells.

Allicin is not bioavailable and will not get absorbed in the blood, even after ingesting large amounts of allicin.

Allicine is de voornaamste thiosulfinaat-verbinding in knoflook (Allium sativum). Allicine is verantwoordelijk voor de karakteristieke (en onaangename) geur van knoflook.

Allicine wordt door de plant gebruikt als beschermingsmechanisme. Wanneer de knoflookplant wordt aangevallen of wordt beschadigd, wordt door een enzymatische reactie allicine gevormd. Het enzym aliinase zet daarbij alliine om tot allicine, wat giftig is voor insecten en micro-organismen.

Het antimicrobiële effect van allicine berust op het feit dat het reageert met de thiol groepen in diverse enzymen, die daardoor onwerkzaam worden.

Allicine is er ook voor verantwoordelijk dat in gebieden met een hoge knoflook consumptie minder maagzweren voorkomen. Onderzoek heeft uitgewezen dat allicine werkt tegen de bacterie Helicobacter pylori, die verantwoordelijk is voor een groot percentage van maagzweren.

Alizarine of alizarinerood is een rood pigment dat oorspronkelijk uit de wortels van meekrap (Rubia tinctorum) gewonnen werd, maar tegenwoordig vooral synthesisch gemaakt wordt. Alizarine is bijzonder geschikt voor het verven van textiel en leer. De kleur wordt ook wel kraplak genoemd.

Meekrap

Meekrap werd als verfstof al gecultiveerd in de klassieke oudheid, met name in Azië en Egypte, waar het reeds in 1500 voor Chr. is aangetroffen. Het is een van de meest stabiele natuurlijke kleurstoffen. Met meekrapwortel gekleurde textiel is dan ook aangetroffen in bijvoorbeeld het graf van Toetanchamon, in de ruïnes van Pompeii en in het oude Corinthië. In de Middeleeuwen werd de kweek van meekrap gestimuleerd door Karel de Grote. Het groeide vooral goed in de zanderige bodem van Nederland, met name in Zeeland, en werd daar ook voor de lokale economie erg belangrijk. Ook in het aangrenzende Bergen op Zoom was een belangrijke industrie. De stad ontleent hier bijvoorbeeld haar carnevalleske naam Krabbegat aan en ook in de Blauwe Handstraat waren ateliers gevestigd.

Meekrap werd waarschijnlijk al in de 12e eeuw in Zeeland verbouwd. Het gewas werd twee of drie jaar na de aanplant geoogst. De plant heeft dikke wortelstokken en dunne bijwortels. Deze laatste bevatten de grondstof van de kleur. De wortels werden gedroogd in een droogoven en daarna verpulverd. Het poeder kon als verfstof worden gebruikt. De ovens, meestoven genoemd, waren een eerste vorm van een coöperatie, waarvan de boeren gezamenlijk gebruik maakten. Na de ontdekking van synthetisch alizarine ging de meekrapteelt ten onder. In Zeeland herinneren straatnamen aan dit ooit voor het gebied zo belangrijke product.

In 1804 ontdekte de Engels verfmaker George Field dat de kleur van meekrap stabieler werd door een behandeling met aluin. Hierdoor werd het een vast en onoplosbaar pigment, met een meer permanente kleur. Door toevoeging van metaalzouten ontdekte men in de jaren daarna dat er diverse andere kleuren van konden worden gemaakt.

Synthetische alizarine

In 1826 ontdekte de Franse chemicus Pierre-Jean Robiquet dat meekrapwortels twee kleurstoffen bevatten, namelijk het rode alizarine en het snel verblekende purpurine. In 1868 werd alizarine de eerste synthetische gemaakte verfstof ooit, toen de Duitse chemici Karl Graebe en Karl Lieberman, in het laboratorium van BASF het maakten uit steenkoolteer, antraceen, door een behandeling met achtereenvolgens kaliumbichromaat en geconcentreerd zwavelzuur. De wereldproductie bedroeg rond 1996 meer dan 7000 ton. De chemische stof heet 1.2 dihydroxyanthraquinone.

Toepassing

De kleurkracht van het pigment is zeer hoog, waardoor het een kleur is die snel in een schilderij bijvoorbeeld gaat overheersen. Door de purperen ondertoon kan het goed met blauwe kleuren tot paars gemengd worden.

In olieverf is de kleur transparant, waardoor de kleur goed voor glaceren gebruikt kan worden. Hoewel de kleur in dunne lagen snel kan verbleken, is het in dikkere lagen goed kleurecht. De kleur droogt erg langzaam in olieverf.

Anethol in etherische olie van Anijs

Anethol is een apolaire aromatische onverzadigde ether, met de formule C10H12O. Anethol geeft de typische smaak aan anijs en venkel. De verbinding is een isomeer van estragol, een verbinding die te vinden is in dragon en basilicum. Anethol heeft een duidelijk zoete smaak en is dertien keer zoeter dan suiker. De smaak wordt prettig ondervonden zelfs in hoge concentraties.

De stof is licht giftig. Het is een chemische uitgangsstof van paramethoxyamphetamine (PMA), wat als XTC verkocht werd en tot enkele sterfgevallen heeft geleid.

Wanneer het alcoholgehalte van een water/ethanol-mengsel hoger dan 45% is, lost anethol op en is de oplossing helder. Door meer water toe te voegen neemt het alcoholpercentage af en de oplosbaarheid van anethol vermindert. Er ontstaat dan een witte suspensie. Anethol is dus oplosbaar in oplossingen met een alchoholpercentage van 45% of hoger.

Anethol, ein Alkyl-phenolether, ist der überwiegende Bestandteil (80 bis 90%) von ätherischen Anis- und Sternanisölen. Auch in anderen Umbelliferenölen, wie z.B. beim Bitterfenchelöl, ist Anethol die Hauptkomponente (ca. 55-75% Anteil, neben Estragol und Fenchon). Süßfenchelöl hat eine ganz ähnliche Zusammensetzung wie das Sternanisöl (80% Anethol neben Estragol, jedoch kaum Fenchon). Anisöl enthält neben Anethol noch Anisaldehyd, Anisketon, Vanillin und Eugenol, welche trotz ihres relativ geringen Anteils eine deutliche geruchliche Abweichung verursachen.

In der Natur kommt hauptsächlich das trans-Isomer von Anethol vor. Als untergeordneter Inhaltsstoff wurde es in Hauhechelwurzeln (Radix Ononidis) sowie in den Pflanzen Magnolia kobus und Artemisia caudata aufgefunden.

Beschreibung:

Bei dem Riechstoff trans-Anethol handelt es sich um farblose Kristalle oder Kristallmassen, die sich in der wärmeren Jahreszeit (oberhalb von ca. 23 °C) zu einem klaren, farblosen bis höchstens schwach gelblichem Öl verflüssigen (genauer Schmelzpunkt 21,4 °C). Der Schmelzpunkt von cis-Anethol liegt bedeutend niedriger (-22 °C), so daß dieses Isomer bei Raumtemperatur immer flüssig vorliegt. Die Substanz ist mischbar mit Aceton, Chloroform, Ethylacetat und Diethylether; sie ist auch leicht löslich in absolutem Ethanol.

Duftcharakteristik / odor profile (trans-A.):

angenehm anisartig, sehr süß, warm-kräuterhaft, würzig. Der Geschmack ist typisch anisartig-süß ("nach Ouzo").

Verwendung

Die angegebenen Verwendungen beziehen sich alle auf das Isomer trans-Anethol.

Anethol kommt als Duft- und Aromastoff, vor allem für Seifen und Mundpflegemittel, zum Einsatz. Wichtig ist daneben die Verwendung in der Spirituosenfabrikation als Aromatiseur für Liköre (Typisch: Pernod, Anisette, Ouzo) und in der Lebensmitteltechnologie (Bestandteil von Aromen, in kleiner Menge z.B. in manchen Fruchtaromamischungen wie Himbeere und Erdbeere, [13]).

In der Pharmazie wird die Substanz auch als aromatischer Wirk- und Hilfsstoff gebraucht (Förderung der Schleimproduktion durch Ausscheidung in die Lungengewebe). Eine antiseptisch-desinfizierende Wirksamkeit wird ebenfalls beschrieben [7].

Wissenschaft: In der Mikroskopie als Einbettungsflüssigkeit benutzt. In der präparativen Chemie zur synthetischen Herstellung von Anisaldehyd (durch gelinde Oxidation mit Kaliumpermanganat oder Kaliumdichromat und 50%iger Schwefelsäure, [5]). Anethol ist in der Ph.Eur.97, Suppl. 99 aufgeführt als Reagenz [4].

Anthocyanins / Anthocyanen

Anthocyanen zijn natuurlijke kleurstoffen die behoren tot de groep van fenolen. Ze zijn rijkelijk aanwezig in groenten, fruit en bloemen. Het is de rode tot blauwe kleur in bijvoorbeeld aardbeien, maar ook in bessen, bonen, petunia's, wijn...

Anthocyanen zijn aan omgevingsfactoren onderhevig zoals licht, zuurstof, zuur, temperatuur, enzymen, ascorbinezuur. Deze factoren hebben meestal een negatieve invloed op de aanwezige anthocyanen zodat de concentratie afneemt met stijgende blootstelling aan de omgeving.

Anthocyanen worden ook als additief toegevoegd aan bijvoorbeeld jam, marmelade en ontbijtgranen. Voor het gebruik van anthocyanen is er geen dagelijkse aanvaardbare maximum dosis vastgelegd.

In de literatuur beweert men dat anthocyanen een positieve invloed hebben op de gezondheid. Ze zouden een zeer belangrijke antioxidant zijn. Ook zouden ze een ontstekingsremmende, anti atherogene en kankerremmende werking hebben. Tenslotte zouden de anthocyanen de nevenwerkingen van diabetes tegen gaan en een positief effect vertonen op de ogen. Veel onderzoek werd gedaan, maar te weinig betrouwbare onderzoek. De dosis die effecten gaf, is te hoog om met een normale consumptie van groenten en fruit te bekomen. Er zijn ook supplementen verkrijgbaar die anthocyanen bevatten, deze bestaan uit een bosbesexract.

Over de kleurstoffen anthocyanen en carotenoïden

Een stof is een kleurstof (pigment) als het een bepaalde kleur licht van het spektrum van zonlicht of een andere wit-licht bron kan absorberen. Zo absorberen bladgroenkorrels rood licht en reflekteren ze dus wit licht minus rood en dat geeft groen.

Er zijn twee typen kleurstoffen: wateroplosbare en niet-water oplosbare (denk maar aan het verschil als je worteltjes kookt en bieten kookt, bij welke gaat het water meekleuren?). De water-oplosbare kleurstoffen heten anthocyanen. De niet water-oplosbare kleurstoffen heten carotenoïden. Er zijn ook nog een paar bijzondere kleurstoffen die niet echt een pigment zijn maar die een kleur geven door de lichtbreking in de struktuur van de moleculen (interferentie). Dit zijn vaak de kleuren in de natuur (bv. van sommige bessen) die een beetje een metaalachtige glans hebben (denk bv. aan de kleur van sommige kevers die metalic groen glanzend kunnen zijn).

Bloemen met dezelfde kleur bevatten meestal ook dezelfde kleurstof(fen).De kleur die wij zien, is vaak een combinatie of mengsel van verschillende kleurstoffen. Door verschillen in het mengsel kan de kleur van bloemen dus verschillend zijn, terwijl er toch dezelfde kleurstoffen in voorkomen.

De kleur van de anthocyanen hangt af van de andere stoffen die in de buurt zitten (metalen, kleurloze flavonoïden, carotenoïden, deze stoffen heten dan co-pigmenten). Bloemen kunnen dus dezelfde anthocyanen bevatten maar door verschillen in co-pigmenten toch een andere kleur hebben. De zuurgraad (pH) van het water in de vacuole van de plantencel (dat is de ruimte waar de oplosbare stoffen zich ophopen) is ook belangrijk voor de kleur die we zien. Verder kan de aanwezigheid van sommige metalen invloed hebben op de kleur die we waarnemen. De anthocyanen behoren tot een grotere groep van 'kleurstoffen' die flavonoïden heten. De meeste flavonoïden zijn voor ons gewoon wit (of kleurloos). Echter, deze pigmenten kunnen wel uv-licht absorberen en voor insekten zien witte bloemen er dus toch gekleurd uit.

Overview:

Anthocyanins are the colorful flavonoids concentrated in brightly colored berries and fruit ("anthos" means "flower" and "cyan" means "blue") being most concentrated in bilberries, blueberries, cranberries, elderberries, purple grapes, red wine and hawthorn berries. Anthocyanin extracts are important for the health of the micro-blood vessel network and are clinically used in this way by medical herbalists and physicians in mainland Europe. Recent studies in the United Kingdom confirm that anthocyanins facilitate the repair of vessel damage responsible for 'small blood vessel permeability' and the related fluid retention caused, including swollen limbs, fingers, breasts and the tissue surrounding the eye area. There is currently little orthodox treatment for this type of fluid retention except diuretics, which are not without side effects and fail to address the underlying cause of the disorder. The ancestral diet was replete with anthocyanins and these antioxidants help to protect the body from harmful free radicals. Bilberry anthocyanins can act to quickly repair and regenerate broken and leaky capillaries and blood vessels within the body. Anthocyanins are also powerful antioxidants that help to protect skin against U.V. rays. Anthocyanins bind to and stabilize collagen and elastin; they stabilize the phospholipids of endothelial cells and increase synthesis of collagen and mucopolysaccharides, which give the arterial walls structural integrity. This strengthening activity could be critical for preventing strokes and cancer. Also, the increased production of collagen and elastin by cells reduces inflammation, which can be beneficial in the treatment of arthritis and asthma. European doctors routinely prescribe anthocyanin extracts before operations to prevent excessive post-operative bleeding. Improved circulation also brings enhanced mental clarity as well. It is well recognized that cranberry and blueberry anthocyanins can effectively treat and prevent mild chronic and recurrent bladder and urinary tract infections; they act to prevent bacteria from adhering to urinary tract walls.

Chemistry:

There are more than 200 different types of anthocyanins that fall into six major groups: delphinidin; cyanidin; petunidin; pelargonidin; peonidin; and malvidin. Kuhnau (1976) describes anthocyanins as water-soluble plant pigments of the 2-phenylbenzophyrylium (flavylium) structure. Different berries have different anthocyanin profiles (HPLC fingerprints) that can be used to identify species and to verify the purity of products including fruit juices (i.e. this is done to verify that cranberry juices sold on the market contain at least 25% real cranberry juice). Different berries also contain different levels of anthocyanins (i.e. dried bilberries contain 0.7% anthocyanins whereas dried commercial blueberries contain only 0.2%). Anthocyanins normally exist bound to sugar molecules (as glycosides) and it is known that without bound sugars (the aglycone) is extremely unstable. The different anthocyanin compounds occur when the different core structures bond with different sugar moieties (glucose < rhamnose < galactose < xylose < arabinose).

For instance, the anthocyanin pigments present in grape-skin extract consist of diglucosides, monoglucosides, acylated monoglucosides, and acylated diglucosides of peonidin, malvidin, cyanidin, petunidin and delphinidin. The amount of each compound varies depending upon the variety of grape and climatic conditions. Bilberries contain a large amount of delphinidin that colors the berries reddish purple. According to Francis (1977), the blue to red colour imparted by the anthocyanins depends largely upon the pH of the medium. The highest quality standardized extracts of bilberries, those used in clinical trials, contain 25% anthocyanins, which corresponds to approximately 36% anthocyanosides (anthocyanins with sugars attached).

Suggested Amount:

Bilberry anthocyanins: Take between 250-1000mg of bilberry extract standardized to contain 25% anthocyanins daily. Alternatively, as a tea, 1-2 tablespoonfuls of bilberry fruit are boiled in water (ca. 150ml) for 10 minutes and passed through a tea strainer while still hot. To treat diarrhea in children or adults, a cup of the freshly prepared tea is drunk cold several times a day.

Cranberry anthocyanins: For treatment of UTIs: The dosage of cranberry juice ranges from between 2 ounces to 16 ounces, based on controlled clinical trials (the lower dosage corresponds to pure cranberry juice and the higher dosage corresponds to cranberry cocktail containing 30% real juice). Powdered extracts of cranberry can be taken correspondingly. For strengthening the vascular system: Take between 250-1000mg of cranberry extract standardized to contain 25% anthocyanins daily. For soothing irritated mucous membranes of the mouth and throat or for treating fever: Place cranberries in cold water for two hours, allowing them to swell and then take 1-2 teaspoonfuls of these berries with some fluid.

Elderberry anthocyanins: For cleansing and fasting, take 1-2 tablespoonfuls of elderberry syrup per day with plenty of liquid (1:10).

Positive Clinical Findings:

A group of Italian researchers showed that a mixture of anthocyanosides from bilberry plus vitamin E stopped the progression of lens clouding in a remarkable 97 percent of people with early-stage cataracts.

Recent Clinical Findings:

A study conducted in Italy found significant therapeutic benefits for using bilberry anthocyanosides to treat fibrocystic breast disease (mastopathy). The study used clinical and instrumental control (echography) of patients before and after at least three months treatment with anthocyanosides. The protocol used was of the prospective and comparative type, whereas follow-up lasted three months. The study was performed in the outpatients' clinic of Breast Physiopathology and Echography organised within the ambit of the services of the Gynecology and Obstetrics Division. In this particular instance both echography and clinical examinations were performed at the same clinic. A total of 257 patients took part in the program of which 35 were excluded since they failed to attend subsequent controls. The study was very well controlled and the results were very encouraging. There was a marked improvement in 75 patients, equivalent to 33.8%, symptoms were reduced in 61 women (27.5%) and disappeared in 14 (6.3%), whereas treatment had no effect in 72 cases (32.4%). In conclusion, therapy with bilberry anthocyanosides for three months in patients with mastodynia, consequent to fibrous mastopathy, was efficacious in reducing symptoms and mammary tension

ANTHOCYANINS: In a review of anthocyanins in fruits, Mazza and Miati (1993) report 300-700 mg/100 g fresh fruit for bilberries, 80-325 for blackberries, 725-1050 in black chokeberry (Aronia melanocarpa) 160 in boysenberries, 250 in black currants, 7-495 for blueberries, 350-450 in cherries, 75-80 for cranberries, 50-400 in hawthorn 30-330 in lingonberry, 1,500 in mountain ash, 10-20 in red currants, 30 to 750 for red grapes 20-60 in red raspberries, 25-180 in saskatoon berry (Amalanchier alnifolia), 140-380 in sea buckthorn 115-225 in whortleberries, (Mazza & Miniati, 1993). Boik (1995) notes that some anthocyanins, as anticoagulants, might prevent blood clots, even strokes. He reports also that they may protect collagen from degradation by inducing cross-linking of collagen fibers, by promoting collagen synthesis, and by preventing enzymatic cleavage of collagen. Anthocyanins inhibit collagenase. By inhibiting collagenase activity, anthocyanins may inhibit invasions by cancer cells. (However, like vitamin C, they may stimulate collagen synthesis and angiogenesis; this latter conflicts with what we read elsewhere, that they might prevent diabetic retinopathy, which at least in some circumstances is caused by new blood vessels proliferation. Boik suggests for arthritis and capillary permeability, doses of 20 to 40 mg pure anthocyanins thrice daily, or 1 20 mg/day. Remeber that 100 g usually calculates to about half a cup. Antrachinonen, laxerende stoffen in Senna

Die Gruppe der Anthrachinone (formal vom Anthrachinon abgeleitet) nimmt in der Pflanzenheilkunde einen wichtigen Platz unter den therapeutisch wirksamen Bestandteilen von Heilpflanzen ein. Sie wirken gegen Verstopfung, da sie den Anteil der Elektrolytmineralien im Darm steigern und den Stuhl weicher machen. Außerdem wirken sie schmerzstillend, entzündungshemmend, gegen Steinbildung und Psoriasis.

Substanzen aus der Gruppe der Anthrachinone

* Aloe-Emodin wirkt antibiotisch und kathartisch.

* Aloetic acid, dt.: Aloesäure hat antibiotische Wirkung.

* Aloin (aus der Aloe vera) steigert die Peristaltik des Dickdarms und wirkt daher leicht kathartisch (abführend), außerdem emetisch (Erbrechen erregend). Aloin ist nur im Blattgrün enthalten, welches bei der Verarbeitung gewöhnlich vorsichtig entfernt wird, so dass es nicht in Aloe vera-Produkte zur inneren Anwendung gelangen sollte.

Warnung: Über einen längeren Zeitraum und in hoher Dosis eingenommen können Anthrachinone eine Gewöhnung herbeiführen und die Darmwand bleibend schädigen! Ein überhöhter Dauergebrauch sollte vermieden werden, da es zur vermehrten Kalium-Ausscheidung kommt. Ein niedriger Kaliumspiegel führt wiederum zur Verstopfung (Obstipation).

Anthrachinone sollten am besten nur in Ausnahmefällen eingenommen werden.

- nach bestimmten Operationen, wenn der Darm geschont werden soll oder wenn die Belastung einer frischen Wunde durch Druck verhindert werden soll.

Antrachinonglycosiden (Sennosiden)

Contactlaxans, in de dunne darm splitsen deze zich in antrachinonen, die laxerend werken door directe prikkeling van het colonslijmvlies. Werking: na 6-12 uur.

Contra-indicaties

Plotseling optredende buikpijn (appendicitis, ileus). Obstructie van de darm. Verstoring van de elektrolytenbalans (m.n. hypokaliëmie). Hoge leeftijd. Sterke lichamelijke verzwakking.

Zwangerschap/Lactatie

Dit geneesmiddel kan, voor zover bekend zonder gevaar voor de vrucht, overeenkomstig het voorschrift worden gebruikt tijdens zwangerschap.

Antrachinonen gaan, bij gebruik van hoge doses, over in de moedermelk en kunnen diarree bij de zuigeling veroorzaken; niet gebruiken tijdens lactatie.

Bijwerkingen

Buikkrampen. Prikkeling van het darmslijmvlies. Elektrolytenverlies. Meteorisme. Misselijkheid. Collaps. Zelden allergische of anafylactische reacties.

Langdurig gebruik kan leiden tot waterige diarree met excessief verlies van water en elektrolyten (vooral kalium), spierzwakte, gewichtsverlies, albuminurie, beschadiging van de plexus myentericus, pigmentering van het colonslijmvlies (melanosis coli).

Interacties

Het kaliumverlies veroorzaakt door andere geneesmiddelen, kan worden versterkt. Bij langdurig gebruik kan de werking van hartglycosiden worden versterkt door optreden van kaliumverlies.

Apigenin is a flavone belonging to the broader family of flavonoids. Apigenin is found in Matricaria recutita and Euphrasia officinalis and other members of the Apiaceae family, and is most known for its antinflammatory and antioxidant action. Apiol is a phenol derived from phenylpropane found in many common spices. Apiol has been shown to have antioxidant properties and is abundant in the oils of Celery (Apium graveolens) and Parsley (Petroselinum crispum).

Organic chemical compound, also known as parsley apiol, apiole or parsley camphor. It is found in parsley seeds and the essential oil of parsley. Heinrich Christoph Link, an apothecary in Leipzig, discovered the substance in 1715 as greenish crystals reduced by steam from oil of parsley. In 1855 Joret and Homolle discovered that apiol was an effective treatment of amenorrea or lack of menstruation.

In medicine it has been used, as essential oil or in purified form, for the treatment of menstruation disorders. It is an irritant and in high doses it is toxic and can cause liver and kidney damage.

Hippocrates wrote about parsley as a herb to cause an abortion. This effect was caused by the apiol.

Apiol was used by women in the Middle Ages to terminate pregnancies. [citationneeded] Its use was widespread in the USA, often as ergoapiol or apergol, until a highly toxic adulterated product containing apiol and tri-orthocresyl phosphate (also famous as the adulterant added to Jamaican ginger) was introduced on the American market.

The toxic effects of pure crystalline apiol are disputed. It causes a "relatively safe abortion" in pregnant women if taken in small quantities. It also restores the cycle of menstruation. A larger dose does not cause an abortion, it causes nausea and damages the liver and kidneys. [citation needed]

Now that other methods of abortion are available apiol is almost forgotten in the West, but it is still produced and is used in the Middle East. [citation needed]

The name apiol is also used for other closely related compounds, found in dill (dillapiole, 1-allyl-2,3-dimethoxy-4,5-methylenedioxybenzene) and in fennel roots.

Arbutine, fenolglycoside in Beredruif

Description

Arbutin is a phenolic glycoside found most notably in Bearberry (Arctostaphylos uva ursi). Arbutin is known to have a potent antibacterial action but requires alkaline urine to allow the antiseptic action to occur. After hydrolysis, arbutin releases a diphenol which oxidises to hydroquinone. While arbutin itself has not been shown to be mutagenic, hydroquinone can be mutagenic and are known to inhibit melanin synthesis. Therefore, as a cautionary measure, arbutin containing plants are not suitable for long term use, in pregnancy or lactation.

Asaron, geurstof in Kalmoeswortel

ist ein Pflanzeninhaltsstoff, der in Kalmusöl enthalten ist, das aus der Pflanze Acorus calamus (deutscher Name: Deutscher Ingwer) gewonnen wird.

Als Reinsubstanz ist Asaron als giftig (kanzerogen) eingestuft, ist aber durch den niedrigen Anteil in Kalmusölen der Grund für deren Verwendung für Heil- und Aromazwecke als auch in der Parfümherstellung. Weil der Stoff zum Niesen reizt, ist er auch Bestandteil des Schneeberger Schnupftabaks.

Vorkommen:

Der Phenylpropan-Abkömmling a-Asaron kommt in der Natur vor, so z.B. im ätherischen Öl der Europäischen Haselwurz (auch Brechwurz genannt, bot. Asarum europaeum L.). Die Konzentration ist in diesem Öl recht hoch, sie beträgt zwischen 30 und 50%.

a-Asaron ist (zusammen mit dem stereoisomeren b-Asaron) auch einer der Hauptinhaltsstoffe des indischen und russischen Kalmusöls von Acorus calamus L. var. spurius (SCHOTT) ENGL. Nordamerikanische und europäische Kalmusöle der Varietäten americanus und calamus weisen dagegen einen weit geringeren Anteil an Asaronen auf. Die Asarone sind in den ätherischen Ölen oft vergesellschaftet mit Elemicin bzw. Isoelemicin.

Beschreibung:

Die Substanz kristallisiert in farblosen, lichtempfindlichen Nadeln, welche in hochprozentigem Ethanol und in organischen Lösungsmitteln (wie z.B. Diethylether, Chloroform, Eisessig und Petroleumbenzin) löslich sind. In Wasser ist a-Asaron praktisch unlöslich. Der Schmelzpunkt liegt bei 62 bis 63 °C.

Sensorische Eigenschaften:

Der Geschmack soll pfefferartig sein [5]. Es handelt sich bei Asaron - wie bei den verwandten Stoffen Myristicin, Elemicin und Safrol - um einen Scharfstoff des o-Methoxy(Methyl)-phenol-Typs (Einordnung nach H. WAGNER).

Vorkommen:Acorus calamus;

Wirkungen:Die Rauschdroge hat erwiesenermassen krebserregende Wirkung.

In hoeherer Konzentration berauschende Wirkung auf den Menschen. Vermutlich wird Asaron im Metabolismus in ein psychedelisches Phenethylamin (TMA 2) umgewandelt, das in kleinen Mengen den Kreislauf stimuliert und das Nervensystem staerkt. TMA-2 hat aehnliche Wirkungen wie Meskalin, vermutet der Hamburger Ethnologe C. Rätsch.

Inhaltsstoffe Acorus

Ätherisches Öl: 2-6 (-9) %: Phenylpropane (alpha-Asaron, beta-Asaron = cis-Isoasaron) Monoterpene ([Z,Z]-4,7-Decadienal, Träger des Kalmusaromas), Sesquiterpenkohlenwasserstoffe (beta-Caryophyllen, Humulen u. a.) und Sesquiterpenketone (thermolabiles Acorogermacron, als Artefakte entstehen Shyobunone, Acoron u. a.). Die Zusammensetzung des Öls und der Ölgehalt der Droge hängen von der Chromosomenzahl und damit von der Herkunft der Pflanze ab. Cis-Isoasaron (beta-Asaron) ist oft neben Iso-Eugenolmethylether Hauptkomponente (bis 80 %). Als charakteristisch für das Kalmusöl der diploiden und triploiden Varietäten können die Sesquiterpene vom Typus des Acorogermacrons und des Acorons angesehen werden.

Verwendeter Pflanzenteil:Von Wurzeln und Blattresten befreiter Wurzelstock (Radix Calami).

Offizinell:Radix Calami: Gehalt an ätherischem Öl mind. 2,0 %, Gehalt an cis-Isoasaron höchstens 0,50 %.

Species amaricantes: je 20 Teile Wermut, Tausendguldenkraut und Bitterorangenschale, je 10 Teile Bitterkleeblatt, Kalmuswurzel, Enzianwurzel und Ceylonzimtrinde.

Species carminativae: je 25 Teile Pfefferminzblatt, Kamillenblüte, Kalmuswurzel und Kümmel.

Therapeutisch relevante Wirkungen:Sekretionsfördernde Wirkung: Kalmuszubereitungen regen aufgrund ihres Gehaltes an Bitterstoffen und ätherischem Öl reflektorisch die Magensaftsekretion (vermutlich auch die Gallensaftsekretion) an.

Weitere Wirkungen:Kalmusöl wirkt als Zusatz in Einreibungen erfrischend und anregend und wird beim varikösen Symptomenkomplex eingesetzt.

Unerwünschte Wirkungen:Chronische Toxizitätsprüfungen an der Ratte mit beta-Asaron-reichem Kalmusöl führten nach einer Applikationsdauer von ca. 60 Wochen zu Tumoren im Zwölffingerdarmbereich. In den USA wurde deshalb jede Art der Anwendung von Kalmus verboten, obwohl gerade die in Nordamerika wachsende diploide Kalmussippe beta-Asaron-freies ätherisches Öl aufweist. Demgegenüber legen die europäischen Arzneibücher und Aromaverordnungen Grenzwerte für den Gehalt von beta-Asaron in der Droge bzw. in Lebensmitteln fest, um so ein eventuelles Gesundheitsrisiko auszuschließen. beta-Asaron besitzt im Vergleich zu anderen Naturstoffen (z. B. Aristolochiasäure) nur eine geringe kanzerogene Wirkungsstärke. Darüber hinaus dürfte auch die für die kanzerogene Wirkung notwendige Metabolisierung beim Menschen anders ablaufen als bei der Ratte. Bisher konnten beim Menschen keine von Kalmus ausgehenden kanzerogenen Effekte beobachtet werden, obwohl in Indien verbreitet der extrem beta-Asaron-reiche Jammu-Kalmus verwendet wird. Literaturhinweise auf den leberkanzerogenen (?!) Effekt einiger "osteuropäischer (?) Arten" beruhen offensichtlich auf einem Zitier- bzw. Abschreibefehler.

Die therapeutische Verwendung beta-Asaron-armer Kalmuswurzeln erscheint daher vertretbar, von einem Dauergebrauch sollte jedoch abgeraten werden, da für genetisch prädisponierte Personen bei lebenslanger Aufnahme von Kalmus ein Krebsrisiko möglich ist.

Adrenocorticotroop hormoon - Corticotropine ACTH

ACTH, internationaal gebruikelijke afkorting van adrenocorticotroop hormoon (v. wetensch. adren = bijnier; Lat. cortex = schors; Gr. trepoo = aandrijven), officiële naam thans corticotropine, ook nog vaak corticotrofine genoemd, een hormoon dat wordt gevormd door de voorkwab van de hypofyse.

ACTH zet de bijnierschors aan tot productie van bepaalde corticosteroïden, m.n. van glucocorticosteroïden (corticosteron en cortisol) en van androgene hormonen van de bijnierschors (het is dus een glandotroop hormoon). Het verlaagt daarbij het gehalte aan vitamine C en aan cholesterol (grondstof voor de productie van bijnierschorshormonen). Daarnaast oefent ACTH invloed uit op de vetsplitsing; het kan uit depotvet vetzuren vrijmaken.

Productie en afgifte:

De productie van ACTH wordt nauwkeurig geregeld. Zij gaat omhoog bij allerlei factoren die het lichaam schade toe kunnen brengen (bijv. ernstige verwondingen of stress). De als gevolg hiervan stijgende productie van bijnierschorshormoon voorkomt overmatige afweerreacties (en beschermt dus het lichaam). De gestegen concentratie van bijnierschorshormoon remt vervolgens de ACTH-productie af.

Overproductie van ACTH is de oorzaak van sommige vormen van het syndroom van Cushing.

Werkingsmechanisme:

Vele hormonen oefenen hun werking uit via hetzelfde basismechanisme: interactie van het hormoon vindt plaats via specifieke receptoren op of in de celmembraan en deze interactie leidt tot activering van het enzym adenylase-cyclase; dit enzym katalyseert de omzetting van ATP in cAMP plus pyrofosfaat. cAMP zet een serie van enzymatische reacties in gang die leiden tot het hormoonspecifieke effect. Voor zover men thans weet geldt dit ook voor ACTH.

Medisch gebruik:

Als geneesmiddel wordt ACTH weinig gebruikt; het is mogelijk het al dan niet functioneren van de bijnierschors vast te stellen door middel van ACTH-injecties.

Structuur:

Corticotropine werd voor het eerst uit varkenshypofyse geïsoleerd door W.F. White (1953); de structuur vond Bell c.s. (1955). ACTH heeft een moleculairgewicht van ca. 4500 en is een peptide, bestaande uit 39 aminozuren; het is gesynthetiseerd (1963). De structuur van corticotropine van de mens is in 1961 opgehelderd door T.H. Lee, A.B. Lerner en V. Buettner-Janusch.

Brokstukken van het ACTH-molecule blijken, bij ratten toegediend, invloed te kunnen uitoefenen op het herinneringsgedrag of de motivatie en op de herseneiwitten.

Atropine, giftig alcaloïde uit Wolfskers

alcaloïde* door extractie verkregen uit Wolfskers (Atropa bella-donna). De bessen hiervan zijn erg giftig; een enkele bes kan soms voldoende zijn om een kind te doden.

Atropine valt farmacologisch gezien onder de parasympaticolytica en heeft vele effecten op het organisme. Hierom is het in veel toepassingen door allerlei afgeleide preparaten verdrongen. Atropine wordt in de moderne geneeskunde echter nog steeds toegepast, met name bij hartritmestoornissen waarbij het hart te langzaam klopt, in noodsituaties bijvoorbeeld bij reanimaties. De gebruikte dosering is hierbij - afhankelijk van de indicatie - fracties van een milligram tot maximaal 4 milligram. Deze bolus kan zowel intraveneus als intramusculair gegeven worden, waarbij de intraveneuze toedieningsvorm binnen enkele seconden tot minuten zijn maximale effect al bereikt, in tegenstelling tot intramusculaire toediening. Over het gebruik binnen een reanimatiesetting zijn de meningen verdeeld over dosering; intermitterend of de maximale dosering in één keer.

Ook vindt atropine nog wel toepassing in oogdruppels als middel om de pupil te verwijden en de accommodatie van de ooglens te verlammen. Overigens houdt dit effect zeer lang aan (tot enige dagen), wat voor de gebruiker vaak een hinderlijke lichtschuwheid en onscherp zien oplevert; meestal worden in de oogheelkunde voor dit doel korter werkende preparaten gebruikt.

Van het effect op de ogen komt ook de wetenschappelijke naam: bella donna = Italiaans voor 'mooie vrouw'. In de oudheid gebruikten vrouwen het sap van de bessen om hun pupil te verwijden.

Overview:

Azulene is a prime component of the essential oil of chamomile flowers, Matricaria recutita L. [Fam. Asteraceae] and related plant species. Products containing azulene generally also contain the other characteristic components of chamomile's essential oil and are gaining popularity for improving leg vein health. Studies have shown that an azulene-based synthetic drug has beneficial cardiovascular effects and may be helpful in treating arrhythmias and for relaxing venous tissues thereby improving circulation. It is thought that this drug may block both Na+ and Ca2+ channels expressed in cardiac and vascular smooth muscles. The multiple ion channel blocking effects of these azulene-based drugs are thought to be largely responsible for the antiarrhythmic and vasorelaxant actions. Azulene extracts used in skin creams for reducing skin puffiness and wrinkles have also gained popularity in the health industry. However, studies have shown that azulene can be photo-reactive following irradiation with UVA and/or visible light, causing a mutagenicity 4-5-fold higher than the spontaneous background mutation. Therefore, it is not recommended to use skin creams standardized for azulene as they may cause skin irritation and mutations. Traditionally, the essential oil of chamomile has been used to help keep bunions under control and to treat Carpel Tunnel Syndrome, sciatica and bone and joint problems through its antinflammatory properties. The German Commission E monograph notes that the oil extract of Chamomile is antiphlogistic (anti-inflammatory and anti-fever) and studies have verified that several compounds contribute to this activity including: bisabolol, chamazulene, matricin and the spiro-ethers. Chamomile tea is also used to prevent and soothe gastrointestinal ulcers. One German synonym for chamomile, translated as 'roll-boil', reflects this traditional usage. To soothe ulcers, a concentrated warm tea of chamomile was drunk while lying down and slowly rolling onto all sides of the body to coat all the inner membranes with the tea.

Chemistry:

Azulene is a prime component of the essential oil of chamomile flowers. Chamomile flowers contain: From 0.3-1.5% essential oil containing azulene, (-)-alpha bisabolol, bisabolol oxides A, B, and C, bisabolone oxide, chamazulene, chamamviolin, spathulenol, cis- and trans-enyne dicyclo ethers (spiro-ether, polyacetylenes) as the principal components.

Suggested Amount:

Chamomile extracts standardized for azulene content are generally taken internally as an herbal product for treating circulatory conditions and are also used in creams for treating dry, puffy skin or wrinkles. However, it is not recommended to use creams standardized for high azulene content as these may cause skin irritation and mutations when exposed to light. The content of azulene in products for internal use should be at least equal to that recommended by the German Pharmacopoeia. German authorities recommend using a heaped tablespoon (ca. 3g) of the flowers for every 150ml of water. It is recommended that boiling water be poured over the flowers and after 5-10 minutes strained. For inflammation of the mucous membranes of the mouth and throat, a freshly prepared cup of tea is used as a gargle or wash. Externally: a 3-10% infusion is used for poultices and rinses; as a bath additive, 50 grams of flowers are used per 10 liters of water.

Contraindications:

Persons who are allergic to daisy family plants [Fam. Asteraceae] may experience allergy symptoms to chamomile. Creams standardized for high azulene content may cause skin irritation and mutations when exposed to light.

The photomutagenicity of the popular skin conditioning agents azulene and guaiazulene were tested in Salmonella typhimurium TA98, TA100 and TA102. Following irradiation with UVA and/or visible light, both azulene and guaiazulene exhibited mutagenicity 4-5-fold higher than the spontaneous background mutation. In contrary, naphthalene, a structural isomer of azulene, was not photomutagenic under the same conditions. Azulene was photomutagenic when irradiated with UVA light alone, visible light alone, or a combination of UVA and visible light. Azulene and guaiazulene are not mutagenic when the experiment is conducted with the exclusion of light. Therefore, extreme care must be taken when using cosmetic products with azulene/guaiazulene as ingredients since after applying these products on the skin, exposure to sunlight is inevitable.

HNS-32 [N(1),N(1)-dimethyl-N(2)-(2-pyridylmethyl)-5-isopropyl-3,8-dimethylazulene-1- carboxamidine] (CAS Registry Number: 186086-10-2) is a newly synthesized azulene derivative. Computer simulation showed that its three dimensional structure is similar to that of the class Ib antiarrhythmic drugs, e.g., lidocaine or mexiletine. HNS-32 potently suppressed ventricular arrhythmias induced by ischemia due to coronary ligation and/or ischemia-reperfusion in dogs and rats. In the isolated dog and guinea pig cardiac tissues, HNS-32 had negative inotropic and chronotropic actions, prolonged atrial-His and His-ventricular conduction time and increased coronary blood flow. In the isolated guinea pig ventricular papillary muscle, HNS-32 decreased maximal rate of action potential upstroke (Vmax) and shortened action potential duration (APD). These findings suggest that HNS-32 inhibits inward Na+ and Ca2+ channel currents. In the isolated pig coronary and rabbit conduit arteries, HNS-32 inhibited both Ca2+ channel-dependent and -independent contractions induced by a wide variety of chemical stimuli. HNS-32 is a potent inhibitor of protein kinase C (PKC)-mediated constriction of cerebral arteries. It is likely to block both, Na+ and Ca2+ channels expressed in cardiac and vascular smooth muscles. These multiple ion channel blocking effects are largely responsible for the antiarrhythmic and vasorelaxant actions of HNS-32. This drug may represent a novel approach to the treatment of arrhythmias.

The azulene-1-carboxamidine derivative N1,N1-Dimethyl-N2-(2- pyridylmethyl)-5-isopropyl-3,8-dimethyl-azulene-1-carboxamidine (CAS 186086-10-2, HNS-32) is a newly synthesized compound. In the present study, direct cardiovascular effects of HNS-32 were assessed using the canine isolated, blood-perfused sinoatrial node, papillary muscle and atrioventricular node preparations, while the antiarrhythmic action was examined using the canine two-stage coronary ligation-induced arrhythmia model. Intracoronary administration of HNS-32 (1-300 micrograms) suppressed the sinus nodal automaticity and ventricular contractile force, while it increased the atrio-His and His-ventricular conduction time as well as the coronary blood flow. Intravenous administration of HNS-32 (5 mg/kg) suppressed the ventricular arrhythmia for approximately 30 min. Since HNS-32 possesses multiple cardiac direct effects which are unique compared with well-established antiarrhythmic drugs, it may become a leading compound in the search for novel antiarrhythmic agents.
